Targeting of the epidermal growth factor receptor (EGFR) has become an established antitumor strategy with anti-EGFR antibodies approved for clinical use or in late stages of development. Postulation of antibody effector mechanisms has been based on in vivo or cell studies. These need to be complemented by an understanding of antibody/EGFR interactions on the molecular level.
