As the endoplasmic reticulum paralog of the cytosolic HSP90, gp96 (grp94, HSP90b1) is an essential molecular chaperone for Toll-like receptors (TLRs) and integrins. Despite structural advancements and intense interests in drug development targeting the HSP90 family, little is known about the structural and functional regulation of gp96 or the mechanism of its action in client folding and maturation. Herein, we provide genetic and biochemical evidence to demonstrate definitively that N-linked glycosylation and disulfide-bond formation of gp96 do not affect its chaperone function.
