Changes in sugar residues of glycoconjugates of chicken salivary lingual glands during development and growth

En vista del escaso conocimiento sobre las secreciones de las glándulas salivales menores de las aves, el propósito del presente trabajo fue investigar la naturaleza y variaciones de la fracción hidrocarbonada en glándulas linguales del pollo durante su desarrollo embrionario y posnatal. Lenguas de embriones de 9 a 19 días, pollos recién nacidos y adultos se procesaron para PAS, Alcian blue y lectinhistoquímica. Los esbozos glandulares aparecieron en embriones de 9 días. Desde los 15 días apareció una secreción PAS positiva y alcianófila a pH 2.5, en glándulas linguales anteriores y posteriores. Desde los 19 días los glicosaminoglicanos sulfatados se localizaron en las células mucosas de las glándulas posteriores y aumentaron hasta la edad adulta. Por el contrario, los glicosaminoglicanos no sulfatados predominaron en las glándulas anteriores. La afinidad variable de estas glándulas con las diferentes lectinas dependió del tipo de lectina, edad del ave y fracción glandular. Nuestros resultados indican que: a) Los cambios histoquímicos se relacionan con el desarrollo y la maduración celular, cumpliendo probablemente algunos azúcares un rol en la inducción y regulación en las primeras etapas de la diferenciación glandular. b) La heterogeneidad del contenido de azúcares en las glándulas linguales anteriores y posteriores indicaría que cumplen diferentes funciones.Taken into account the scarce data available regarding the secretions of avian minor salivary glands, we undertook to study the nature and the variations of the carbohydrates residues of glycoconjugates in the chicken lingual glands during embryonic life and postnatal growth. Tongues of 9 to 19 day chick embryos, post-hatched and adult chickens were analyzed by H/E, PAS, Alcian blue and lectin histochemistry. At the 9 th day of incubation, primordia of the lingual glands were observed. From the 15 th day, an appreciable degree of reactivity with PAS, Alcian blue and lectin histochemistry was detected at the cells of anterior and posterior lingual glands. From the 19 th day of incubation sulfated glycosaminoglycans were located in mucous cells of the posterior lingual glands, which showed an increase in stainability till adult chickens. On the contrary, nonsulfated glycosaminoglycan were predominant in the anterior lingual glands. Lectin staining depended on the lectin type that was applied, but also on the age and glandular part both in anterior and posterior lingual glands. The results showed that the degree of histochemical changes depends upon the stage of differentiation and maturation of the glands; some sugar residues seemed to play a role inducing and regulating the first differentiate steps of the glands. Differences in sugar content, in both anterior and posterior lingual glands would indicate their various roles.Facultad de Ciencias Veterinaria
