The DNA-binding protein Spo0A is the master regulator for initiation of sporulation in Bacillus subtilis, which is also the case in Clostridium acetobutylicum. Radioactive primer extension experiments revealed two different transcription start poits for spo0A in C. acetobutylicum. Like in B. subtilis the activity of the two start points are depending on the growth phase. The signal S1 allowed to identify a clear sA-dependent -10- and -35-sequence. A sH-dependent -10- and -35-promoter sequence can be postulated from the other signal S3. But in C. acetobutylicum there does not exist the known promoter switching from B. subtilis.
