Reaction of sera of cultivable fishes in India with monoclonal antibody to immunoglobulin of Rohu (<i>Labeo rohita</i>) in immunodot

292-295Monoclonal antibodies (MAbs) can be employed for quantification of serum and mucus Immunoglobulin (Ig) of related fishes. Evaluation of cross reactivity of MAb with various antigens is an important aspect in determining the specificity of the MAb. In the present study, an immunodot assay was employed for cross reactivity studies of anti-rohu immunoglobulin MAbs to an array of fish sera instead of the usually followed techniques such as ELISA and Western blot. The Immunodot revealed that the MAb F4G12 reacted with epitopes of Ig of only Cyprinid fishes while F2D9 recognised an epitope present in all the Cyprinids tested, except common carp. Similarly, among the two MAbs, MAb F4G12 was found to be reacting with both sera and mucus of rohu. These results indicate that immunodot can be used for cross reactivity studies of MAbs with a large number of antigens rapidly and cost effectively
