Study of selenium's effects relative to the production of reactive oxygen species and selenoproteins in genetically modified and unmodified Arabidopsis thaliana plants

Orientador: Marco Aurélio Zezzi ArrudaDissertação (mestrado) - Universidade Estadual de Campinas, Instituto de QuímicaResumo: Com o objetivo de avaliar os efeitos do selênio (Se) e/ou da modificação genética no metabolismo das plantas são realizados estudos de estresse oxidativo, resistência de acumulação de Se e análises metalômicas utilizando folhas de Arabidopsis thaliana modificadas geneticamente (MG) e não modificadas geneticamente (N-MG), e cultivadas em casa de vegetação com irrigação de selenito de sódio. As plantas MG e N-MG estudadas apresentam algumas diferenças devido à presença do selenito. As plantas N-MG, quando irrigadas com a dose superior de Se (308 µg), acumulam 17 % menos Se e sofrem 5 % a mais de redução no crescimento, refletida na menor quantidade de massa orgânica fresca das folhas, quando comparadas com as plantas MG. Além disso, ao comparar as plantas N-MG controle e as tratadas com Se, é observada uma redução até de um 10% na atividade da SOD e um aumento do até 162 % na concentração de MDA. Entretanto, as folhas MG apresentam um 48 % de aumento na atividade da SOD e na produção de H2O2 na dose superior de Se, enquanto que a concentração de MDA diminui em 11 %. Por sua vez, a concentração de proteínas solúveis no extrato das folhas N-MG é maior em comparação às MG (ca. 33 %), e não apresentam diferenças na presença do selenito, enquanto que as folhas MG apresentam uma diminuição de 32 % na maior concentração de Se. No sistema SEC-DAD-ICP-MS é possível o monitoramento do aumento nas áreas dos sinais cromatográficos das espécies de Se solúveis em água, nas folhas de A. thaliana N-MG irrigadas com uma solução de selenito de sódio, quando comparadas com as MG. Os resultados dos estudos das folhas de A. thaliana MG e N-MG, sob os diferentes tratamentos, permitem avaliar, tanto a modificação genética, quanto a presença do Se, as quais produzem efeitos sobre o metabolismo da planta, fazendo que ela produza uma maior concentração de ERO. A modificação genética também confere uma maior resistência ao estresse oxidativo produzido pela presença do selenito. Além disso, é possível identificar variações no conjunto de compostos hidrofílicos, como, por exemplo, proteínas, entre as folhas de A. thaliana N-MG e MG controle e irrigadas com uma solução de selenito de sódioAbstract: In order to evaluate the effects of selenium (Se) and/or the genetic modification in plants metabolism, oxidative stress and resistance of Se accumulation studies and also metalomics analysis are performed using, Arabidopsis thaliana leaves, genetically modified (MG) and unmodified (N-MG), in a greenhouse and irrigated with sodium selenite. MG and N-MG plants show some differences due to the presence of selenite. The N-MG plants when irrigated with the highest dose of Se (308 µg), accumulate 17% less Se and had 5% more reduction in their growth (reflected in low amount of fresh organic mass of leaves), comparing with MG plants. In addition, when N-MG control plants are compared with those treated with Se, a reduction of up to 10% in the SOD activity and an increase of 162% in MDA concentration are observed. Moreover, when the highest dose of Se is used, MG leaves show a significant increase (48 %) in SOD activity and production of H2O2, and a decrease of 11 % on the MDA concentration. However, the concentration of soluble protein in the extract of N-MG leaves is more than 33% higher compared with MG leaves, and no difference in the presence of selenite, while the MG leaves show a decrease of 32% in the highest concentration of Se. The SEC-DAD-ICP-MS system, allows the monitoring of the increase in the chromatographic peaks of selenium species soluble in water in N-MG A. thaliana leaves, irrigated with sodium selenite solution when it is compared with MG leaves data. Results of studies of MG and N-MG A. thaliana leaves, under different treatment conditions, allow to evaluate not only the genetic modification, but also the presence of selenium, which both effects play an important role in plant metabolism, such as a higher production of reactive oxygen species. The genetic modification also confers an increased on the plants resistance against the oxidative stress produced by the presence of selenite.
