Transderrnal therapeutic systerns. II. Rol of skin in drug absorption

En este trabajo se analizan las distintas barreras difusionales que deben atravesar las molecúlas de una droga cuando son absorbidas a través de la piel hasta llegar a la circulación sanguínea. La principal barrera de penetración reside en el stratum corneum. Cuando una droga es liberada hacia la piel por un sistema terapéutico transdérmico (S.T.T.) a una velocidad menor que la velocidad de penetración, el sistema juega el rol controlante en la concentración sanguínea. Se consideran sistemas cuyas cinéticas de liberación son de orden cero, uno y 1/2. Se analizan los factores que afectan la velocidad de penetración de una droga a través de la piel.The diffusional resistances that the drug molecules have to overcome during the course of penetration across the skin tissue and the subsequent uptake by the capillary network for transport to the general circulation are described. The mean diffusional barrier has been found to resid in the stratum corneum cell layers. If the drug is delivered to the skin via a transdermal therapeutic system (T.T.S.) at a lower releasing rate than the rate of percutaneous absorption, the process of drug release will play the rate-controlling role to the blood levels of drugs. The zero, first and 112 order rate constant of the drug delivery process from a T.T.S. are considered.
