PENGGUNAAN KITOSAN TERADIASI SINAR GAMMA75 kGy DALAM FORMULASI NANOPARTIKEL GAMAVUTON-0 DENGAN METODE GELASI IONIK

Chitosan is a natural biopolymer which has been widely used in nanoparticle formulation because it has many advantages, such as mucoadhesive, biocompatible, biodegradable, non-toxic, and low immunogenicity levels in the human body. However, chitosan has a heavy molecular weight, high viscosity, and low solubility, which is limiting its application. Strategy to lower the molecular weight of chitosan was by using 75 kGy gamma radiations. The irradiated chitosan was used to formulate nanoparticle with gamavuton-0 (GVT-0) as an active compound using ionic gelation method. The aim of this study was to know the effect of using 75 kGy irradiated chitosan on characteristic of GVT-0 nanoparticle. Preparation was done by using ionic gelation method in pH 4.00. GVT-0 nanoparticle was made by mixing GVT-0 which had partially negative charge and 75 kGy irradiated chitosan which had positive charge, so that nanoparticle was formed spontaneously by electrostatic interaction. Then, the GVT-0 nanoparticle was stabilized by adding tripolyphosphate as a cross linker. In order to obtain the optimum formula of GVT-0 nanoparticle, the GVT-0 nanoparticle was characterized such as particle morphology, particle size, polydispersity index, zeta potential, and entrapment efficiency. The result of this study was found that the optimal formula for preparing nanoparticle by mixing 0.02% GVT-0 0.50 mL, 0.04% 75 kGy irradiated chitosan 0.50 mL and 0.001% tripolyphosphate. The optimized GVT-0 nanoparticle exhibited spherical morphology, a mean particle size of 55.6±5.7 nm, polydispersity index of 0.280±0.109, zeta potential of +24.27±1.4 mV and entrapment efficiency of 92.84±0%. Based on this study could conclude that the optimum formula of GVT-0 nanoparticles by using 75 kGy irradiated chitosan had better characteristic than the optimum formula of GVT-0 nanoparticles by using 0 kGy irradiated chitosan
