LEPTIN GENE POLYMORPHISM IN ISCHEMIC STROKE PATIENTS WITH DYSLIPIDEMIA

Background: Ischemic stroke is a heterogenous disorder with various underlying pathophysiology. Genetic studies is done to identify the role of genes as a risk factor of ischemic stroke. Objective: This research identifies the role of leptin gene polymorphism as a risk factor for dyslipidemia and ischemic stroke. Dyslipidemia is one of the mechanisms that may lead to ischemic stroke. Method: Case control study was conducted on 42 cases and 42 controls selected randomly. Cases were samples from ischemic stroke patients in RSUD dr Zainoel Abidin Banda Aceh whereas controls were samples from the healthy population in Banda Aceh. DNA isolations and PCR were done to study the polymorphism of microsatellite alleles of leptin gene in these samples. Linear regression and correlation studies were done to identify the relationship between the variables. Results: Results showed that there is no direct relationship between leptin gene polymorphism, dyslipidemia and ischemic stroke occurrence. Further observations on other contributing variables showed that leptin gene polymorphism influences HDL levels (p=0.002)and HDL/LDL ratio (p=0.018). HDL/LDL ratio influences the occurrence of dyslipidemia by 40.2%. Leptin gene polymorphism affects the levels of HDL (p=0.002) and HDL has a significant relationship to the occurrence of ischemic stroke (p=0.001. Conclusion:. Through indirect relationships, polymorphism of microsatellite alleles of leptin gene is a risk factor of dyslipidemia and ischemic stroke occurrence
