ISOLASI DAN UJI AKTIVITAS ANTIBAKTERI MINYAK ATSIRI DAUN TEKI (Cyperus rotundus)

A research related to isolation of Cyperus rotundus leaves essential oil and antibacterial activity test against Staphylococcus aureus and Escherichia coli has been done. The research aimed to isolate the essential oil of Cyperus rotundus leaves and identify the chemical component contained in there, obtain the essential oil of Cyperus rotundus leaves by the preparatif TLC method and identify the chemical component contained in each fraction, and test antibacterial activity of essential oil of Cyperus rotundus leaves and its fraction. Cyperus rotundus leaves cut into pieces and air-dried. Then, the isolation of essential oil was done for 5 hours by water distillation. Separation of chemical compound isolated from essential oil was conducted using preparatif TLC method and eluent used for separation selected by TLC method. Identification of chemical component of essential oil and its fraction has been done by GC-MS, and its antibacterial activity was tested. Distillation result showed that yield of essential oil using water distillation is 0,05% (v/b dry). Eluent used in the separation of essential oil of Cyperus rotundus leaves by preparative TLC method are n-hexane : diethyl ether in the ratio of 8 : 2 (v/v). Retardation factor of fraction I is 0.03, fraction II is 0.53 and fraction III is 0.84. Antibacterial activity of essential oil obtained from water distillation shows as much as 7.25 mm Minimum Inhibition Diameter (MID) in Staphylococcus aureus and 10.50 mm in Escherichia coli bacteria. Fraction I of essential oil of Cyperus rotundus leaves also shows an antibacterial activity. Antibacterial activity fraction I of essential oil is 8.25 mm of Minimum Inhibition Diameter (MID) in Staphylococcus aureus and 11.75 mm in Escherichia coli bacteria.
