Studies on the effect of metal salts to the formation of amylase by Aspergillus niger

[Author abstract]In this paper Aspergillus niger van Tieghem was used for studies on the effect of metal salts to the formation of liquefying amylase and saccharifying amylase by Fungi. Various salts used in the investigation were CuSO_4, CoCl_2, NiCl_2, MnCl_2, SnCl_2, PbSO_4, ZnSO_4, HgCl_2, BaCl_2 and A1_2(SO_4)_3. These salts were added in the incubating medium at various concentration from 10^ to 10^ M. Aspergillus niger was incubated by the method of shaking culture at 35℃ for 6 days, and the incubating medium was used for the determination of liquefying amylase and saccharifying amylase. In the case of the addition of MnCl_2, Al_2 (S0_4)_3 or CuSO_4 these salts acted effectively to form of liquefying amylase and saccharifying amylase. The effective concentration of MnCl_2 was from 10^ to 10^M, its concentration of A1_2(SO_4)_3 was from 10^ to 10^M and its concentration of CuSO_4 was 10^M. On the other hand, in the case of the addition of HgCl_2 , PbSO_4 or BaCl_2 these salts inhibited to form of each smylase at the range from 10^ to 10^M. And in the case of the addition of CoCl_2, ZnSO_4 or SnCl_2, these salts did not act effectively to form of each amylase at the range from 10^to 10^M. In these results the interaction of metal salts and the formation of liquefying amylase and saccharifying amylase was discussed.[要約]Aspergillus niger van Tieghemによる液化amylase, 糖化amylaseの生成にたいする各種の金属塩頬の影響について検索した｡MnCl_2, Al_2(SO_4)_3, CuSO_4の添加により、液化amylaseおよび糖化amylaseの生成にたいして促進効果が認められ、その濃度はMnCl_2では10^～10^M, Al_2(SO_4)_3では10^～10^M, CuSO_4では10^Mであった。とくにMnCl_2, Al_2(SO_4)_3の添加は酵素の生成に有効であることが認められた。HgCl_2, PbSO_4, BaCl_2の添加の場合には、amylaseの生成は10^Mでも阻害され、HgCl_2では10^～10^M, PbSO_4では10^～10^M, BaCl_2では10^～10^Mで酵素の生成があまり認められず、菌の生育の抑制も著しい｡SnCl_2, CoCl_2, NiCl_2, ZnSO_4では低濃度の10^M, 10^Mにおいてもamylaseの生成にたいし促進効果が認められなかった。Mn^, Al^, Cu^の促進効果は、TCAcycle、炭水化物代謝、アミノ酵代謝を活発にし、その結果として効果があらわれたものと推察した
