Studies on acid-stable Protopectinase from Aspergillus awamori

[Author abstract]Studies on an acid-stable protopectinase (PPase), from Aspergillus awamori IFO 4033,were done. We found that the strain produced extremely acid-stable PPase in a solid-state fermentation using wheat bran as the medium. The enzyme, PPase-AS, was stable at pH 2.0 and 37℃ for 24 h, and was most active at pH 2.0 and 50℃. To characterize this outstanding acid-stable PPase, PPase-AS, comparative studies with the less-acid-stable isozyme, polygalacturonase-X2 (PGase-X2,which loses activity within 4 h at pH 2.0 and 37℃), were conducted. The two PPase genes that encode PPase-AS and PGase-X2 were cloned and characterized. The deduced amino acid sequences of these PPases indicate that PPase-AS contains more acidic amino acids, such as aspartic acid and glutamic acid, than PGase-X2. Considering the relative acidity of these enzymes together with the their deduced amino acid sequences, we speculated that acidic amino acids located on the surface of the enzyme molecule might contribute to the stability of PPase-AS under acidic conditions. Two genes have PacC binding sites on the upstream regions of their open reading frame, and their expressions seem to be purposively regulated by ambient pH. PPase-AS showed pectin-releasing activity toward protopectins from various origins and macerating activity toward various plant tissues. Thus, PPase-AS appears to be favorable for industrial use, as bacterial infection is hindered by the suitable conditions, i.e., extremely low pH, for PPase-AS reactions.[著者抄録]黒麹菌(Aspergillus awamori)が生産する耐酸性プロトペクチナーゼ(PPase)に関する研究を行った。著者らは黒麹菌が小麦フスマを培地とした固相培養により耐酸性PPaseを生産することを見出し, 同酵素の精製および諸性質の検討を行った。本酵素, PPase-ASはpH 2.0,37℃で24時間以上安定であり, 同pH, 50℃で最も強い活性を示した。PPase-ASの特異性を明らかにする目的で非耐酸性アイソザイム, PGase-X2を単離し, 諸性質の検討を行ったが, PGase-X2はpH 2.0,37℃で加温すると, 4時間で完全に活性を失った。これら2種の酵素のpHに対する特性(最適pH, 安定性)は大きく異なり, 等電点にも違いが認められた。PPase-ASの耐酸性機構を明らかにする目的で2種のPPase遺伝子の単離を行った。クローン化した遺伝子の塩基配列より推定されるアミノ酸配列を比較すると, PPase-ASはPGase-X2と比較して酸性アミノ酸残基数が多く, 塩基性アミノ酸残基数が少ない事が見出された。2種の酵素の等電点が異なるという事実より, これらの酸性アミノ酸残基が酵素の分子表面に位置することがPPase-ASの耐酸性に寄与しているものと推測した。また, 2種のPPaseの生産量は培地pHにより転写レベルで制御されている事を明らかにした。これらのPPase遺伝子の上流域にはpHによる転写制御因子(PacC)の結合配列が存在しており, 本菌がPacC類似遺伝子を有していたことから, 両酵素の生産は培地のpHにより合目的に制御されているものと考えた。PPase-ASは様々な植物組織よりペクチンを遊離する活性を有しており, 植物組織を単細胞化する活性を持つ。本酵素の反応最適pHは極度な酸性条件であり, 酵素反応液には殆ど微生物による汚染が起こらないことから, 本酵素を用いたペクチンの抽出や単細胞化食品の生産など食品産業への応用が期待される結果を得た。記事区分：原
