Avaliação da atividade biológica de metabólitos de Lippia alba (Verbenaceae), Plectranthus amboinicus e Plectranthus barbatus (Lamiaceae)

This study aimed to identify substances with antimicrobial, antiparasitic and antitumor activities of extract and essential oil from the leaves of Lippia alba (Mill.) N.E. Brown and evaluate the antimicrobial and antitumor activities of essential oils from leaves of Plectranthus amboinicus (Lour.) Spreng and Plectranthus barbatus Andrews. The major compounds identified in the essential oil of L. alba leaves were monoterpenes nerol/geraniol (27%) and neral/geranial (22%), the ketone, 6-methyl-5- hepten-2-one (12%) and sesquiterpene, (E)-caryophyllene (9%). This oil showed cytotoxic activity in vitro against B16F10 (murine melanoma) with IC50 of 45.82 µg/mL. Furthermore, it was found that the oil has antimicrobial activity in vitro, inhibiting Gram-negative bacteria, except Pseudomonas aeruginosa, Gram-positive bacteria and yeasts. The dichloromethane phase obtained from the methanol extract of the leaves of this species showed activity against Leishmania (Leishmania) amazonensis with IC50 of 50.8 µg/mL and among the active groups of fractions were identified triterpenes mixture of oleanolic and ursolic acids, in addition to the steroid stigmasterol, all described in L. alba for the first time. The essential oil from leaves of P. amboinicus presented as major compounds carvacrol (38%), γ-terpinene (15%), (Z)-caryophyllene (14%) and p-cymene (12%), showing cytotoxic activity against B16F10 with IC50 similar to standard drug, however, twice more specific, in addition, showed antimicrobial activity against S. epidermidis, C. albicans, C. neoformans (serotype D), C. gattii (serotypes B and C) and S. cerevisiae. In turn, essential oil from leaves of P. barbatus presentes as major compounds (Z) -caryophyllene (18%), germacrene -D (17%), viridiflorene (14%), cyclosativene (10%) and α -pinene (9%), and has demonstrated activity against C. neoformans (serotype A) and C. gattii (serotype C).O presente estudo teve como objetivo identificar substâncias com atividades antimicrobiana, antiparasitária e antitumoral a partir do extrato e do óleo essencial das folhas de Lippia alba (Mill.) N. E. Brown, bem como avaliar a atividade antimicrobiana e antitumoral dos óleos essenciais das folhas de Plectranthus amboinicus (Lour.) Spreng e Plectranthus barbatus Andrews. Os compostos majoritários identificados no óleo essencial das folhas de L. alba foram os monoterpenos isômeros nerol/geraniol (27%) e neral/geranial (22%), a cetona, 6- metil-5-hepten-2-ona (12%) e o sesquiterpeno, (E)-cariofileno (9%). Este óleo apresentou atividade citotóxica in vitro frente à B16F10 (melanoma murino) com CI50 semelhante à droga padrão, mas menos tóxico. Além dessa atividade, foi verificado que o óleo possui atividade antimicrobiana in vitro, inibindo bactérias Gramnegativas, exceto Pseudomonas aeruginosa, Gram-positivas e leveduras. A fase diclorometânica obtida a partir do extrato metanólico das folhas dessa espécie apresentou atividade antipromastigota de Leishmania (Leishmania) amazonensis com CI50 de 50,8 µg/mL e dentre os grupos de frações ativos, foram identificados a mistura dos triterpenos ácidos oleanólico e ursólico, além do esteroide estigmasterol, todos descritos em L. alba pela primeira vez. O óleo essencial das folhas de P. amboinicus apresentou como compostos majoritários o carvacrol (38%), γ-terpineno (15%), (Z)-cariofileno (14%) e p-cimeno (12%), demonstrando atividade citotóxica contra B16F10 com CI50 semelhante à droga padrão, porém, duas vezes mais específico, além disso, apresentou atividade antimicrobiana contra S. epidermidis, C. albicans, C. neoformans (sorotipo D), C. gattii (sorotipos B e C) e S. cerevisiae. Por sua vez, o óleo essencial das folhas de P. barbatus apresentou como compostos majoritários (Z)-cariofileno (18%), germacreno-D (17%), viridifloreno (14%), ciclosativeno (10%) e α-pineno (9%), e demonstrou atividade contra Cryptococcus neoformans (sorotipo A) e C. gattii (sorotipo C).Dados abertos - Sucupira - Teses e dissertações (2013 a 2016
