Abstract

African Crop Science Journal, Vol. 9. No. 2 June 2001 Isolation and regeneration of protoplasts from mycelium of Fusarium solani F. ELBOUAMI Code Number: cs01016 ABSTRACT Protoplasts from Fusarium solani wild-type strain were readily obtained using snail or microbial preparations. The maximal yields dependent upon the origin of the lytic preparation, age and amount of mycelium. It also depended upon the nature and molarity of the stabilisers. Optimal production (109 protoplasts per g of fresh weight mycelium) was reached by treating 20 hr.-old cultures with Novozym in the stabiliser 0.6M KCl. According to three different regeneration patterns, up to 30% of the released protoplasts regenerated to mycelium. Key Words: Lytic enzymes, Novozy...
