Isolation and regeneration of protoplasts from mycelium of Fusarium solani

ELBOUAMI Code Number: cs01016 ABSTRACT Protoplasts from Fusarium solani wild-type strain were readily obtained using snail or microbial preparations. The maximal yields dependent upon the origin of the lytic preparation, age and amount of mycelium. It also depended upon the nature and molarity of the stabilisers. Optimal production (109 protoplasts per g of fresh weight mycelium) was reached by treating 20 hr.-old cultures with Novozym in the stabiliser 0.6M KCl. According to three different regeneration patterns, up to 30% of the released protoplasts regenerated to mycelium. Key Words: Lytic enzymes, Novozym, nuclear staining culture RÉSUMÉ Les protoplastes de Fusarium solani issus d'une souche type sauvage étaient facilement obtenus par utilisation des préparations microbiennes ou de l'escargot. Les rendements maximaux dépendaient de l'origine de la préparation lytique, l'âge et la quantité du mycélium de même que la nature et la molarité des stabilisants. La production maximale (109 protoplastes/gr de mycélium frais) était obtenue par traitement des cultures âgées de 20 heures au Novozym, dans un stabilisant de 0.6M KCI. D'après trois modes de régénération, jusqu'à 30% des protoplastes libérés se sont régénérés en mycélium.
