L'un des principaux objets d'étude de la théorie des matrices aléatoires est le spectre de matrices dont les coefficients sont des variables aléatoires et dont la dimension est grande. Dans cette thèse, on s'intéresse à deux questions concernant le spectre de matrices aléatoires : les grandes déviations de la plus grande valeur propre et la convergence de la mesure empirique. En dehors des cas où les coefficients sont à distributions gaussiennes ou à queues lourdes, peu de principes de grandes déviations sont connus en théorie des matrices aléatoires, tant pour la mesure empirique que pour la plus grande valeur propre.
