Etude de l'effet des ultrasons couplés à un pilote de filtration tangentielle sur la clarification du jus de jaca (Artocarpus heterophyllus L)

Jackfruit is an exotic fruit with high nutritional value and significant industrialization potential in the clarified juice (CJ) market. Membrane filtration systems (MFS) make it possible to obtain JC with microbiological stability while retaining their physicochemical (PCP) and nutritional properties. However, problems related to the deposition of solids (clogging) on the membrane surface (Ms) have been reported. The use of Ultrasound (US) coupled to MFS represents an innovative alternative for the reduction of this phenomenon, with the aim of increasing the process performance, but, there isn’t study that has noted their effect on fruit juice clarification, where the main responsible solutes for the clogging are the pectins, however, there isn’t work to date that has been able to highlight them concretely. Studies conducted by LRP and Grenoble ESRF have shown the formation of concentrated layers on Ms and the US effect on MFS using small-angle X-ray scattering (SAXS). In this context, the thesis objective was to study the effect of US coupling to tangential microfiltration (TMF) pilot on process performance, PCP and microbial stability of jackfruit CJ. The work was divided into 5 steps:1) Enzymatic hydrolysis: Six enzymatic treatments (tmt) were applied to the jackfruit pulp (JP), under constant incubation conditions, optimized by the surface response methodology (SRM), where time (t) and concentration (C) were the response of reducing sugars (RS) and viscosity (η).2) Stabilization of Hydrolyzed Juice (HJ) and JP by Spray Drying (SD): Two maltodextrin concentrations (MC) were selected for SD at constant conditions; drying performance (Dp) and rheological behavior (Rb) were defined the best tmt.3) TMF under US (preventive mode): The SD juices were reconstituted and microfiltered in a laboratory cell, and the influence of several parameters, the pore size (0.1 and 0.22 μm, PVDF planes), the tangential flow rate (0, 5 and 0.3 L / min) and the US application (with/without) (20 kHz, 8 W.cm-2) were studied on the flux and PCP; the best tmt was evaluated in a pilot MFS (TAMI 60 cm) with/without US (35 kHz, 27 W.cm-2).4) Demonstration of pectin clogging and US action (corrective mode) / SFM by SAXS: TMF experiments of juice-model (12% sucrose, 0.6% citric acid and 0.75% pectin) were performed from optimal conditions to the ESRF on cells coupling filtration/SAXS. Pectin concentration profiles were highlighted.5) Microbiological analysis and PCP of the CJ in the pilot MFS: CJ with/without US were analyzed to identify the US effect on their quality and microbiological flora.The results: Celluzyme® + Pectinex ultra pulp® enzymes (1%, 3 h) allow a 94.5% reduction in η. The SRM indicates that the decrease in η depends on t, because of the pectin content in the JP (3.2-5.8%), the main responsible for the η. On the contrary, the increase of RS depends on C. For the SD the best tmt for JP and HJ was 25 and 50% of CM, with a Dp of 57 and 74% respectively, without affecting its Rb. In the TMF, the US application on the 0.1 μm membrane doubled the flux at low pressures and the 0.22 μm led to an immediate decrease in flux a low TMP, this one induced by blockage clogging. The PCP show that the US doesn’t affect the treated juice and promotes the release of interest compounds. In the pilot MFS the flux increased in the presence of US, but a reduction of carotenoids and microorganisms of JC was observed in connection with the application mode of the US. The SAXS measurements during the TMF of juice-model revealed for the first time the pectin accumulation at the Ms inducing a decrease in the flux, so they highlighted a synergistic effect between the tangential flow rate and the US for the reduction of concentrated layers.Clarification de jus de fruits, oú les principaux solutés responsables du colmatage sont les pectines mais aucun travail à ce jour n’a pu concrètement le mettre en évidence. Des études menées au LRP et à l’ESRF de Grenoble ont montré la formation des couches concentrées à la Sm et l’effet des US sur SFM en utilisant la diffusion de Rayons X aux petites angles (SAXS). Dans ce contexte, l´objectif de la thèse était étudié l’effet du couplage des US à un pilote de microfiltration tangentielle (MFT) sur la performance du procédé, les PPC et la stabilité Mb du JC de jaca. Le travail a été divisé en 5 étapes :1) Hydrolyse enzymatique: 6 traitements(tmt) enzymatiques ont été appliqués à la pulpe de jaca (PJ), en condition constantes d’incubation, optimisé par la méthodologie de surface de réponse (MSR), où les facteurs temps (t) et concentration (C) ont eu comme réponse les sucres réducteurs (SR) et la viscosité (η). 2) Stabilisation du jus hydrolysé (JH) et de la PJ par séchage par atomisation (SA): 2 concentrations de maltodextrine (CM) ont été sélectionnées pour le SA à conditions constantes; les performances de séchage (Ps) et le comportement rhéologique (Cr) définissent le meilleur tmt. 3) MFT sous US (mode préventif): Les jus SA ont été reconstitués et microfiltrés dans une cellule de laboratoire, et l’influence de la taille des pores (0,1 et 0,22 µm, PVDF planes), le débit tangentiel (0,5 et 0,3 L/min) et l’application des US (avec/sans) (20 kHz, 8 W.cm-2) ont été étudiés sur le flux et le PPC. Le meilleur tmt a été évalué dans un SFM pilote (TAMI 60 cm) avec/sans US (35 kHz, 27 W.cm-2). 4) Mise en évidence du colmatage de la pectine et de l’action des US (mode correctif)/SFM par SAXS : Des expériences de MFT de jus-modèle(12% saccharose, 0.6% acide citrique et 0.75% pectine) ont été effectuées à partir de conditions optimales à l’ESRF sur des cellules couplant filtration/SAXS. Des profils de concentration de pectines ont été mis en évidence.5) Analyse Mb et PPC du JC dans le SFM pilote : les JC avec/sans US ont été analysés pour identifier l’effet des US sur leur qualité et flore Mb.Les résultats : Les enzymes Celluzyme®+Pectinex ultra pulp® (1%, 3 h) permettent une réduction de 94,5% de η. Le MSR indique que la diminution de η dépend de t, en raison de la teneur en pectine dans la PJ (3,2-5,8%), principal responsable de la η. Au contraire, l’augmentation de SR dépend de C. Pour le SA le meilleur tmt pour la PJ et le JH était 25 et 50% de CM, avec une Ps de 57 et 74% respectif, sans affecter son Cr. Lors de la MFT, l'application d’US sur la membrane de 0,1 µm a permis de doubler le flux aux faibles PTM. Sur la membrane 0,22 µm une diminution immédiate du flux a été observée, induite par un colmatage par blocage. Les PPC montrent que les US n'affectent pas le jus traité et favorisent la libération de composés d'intérêt. Dans la SFM sur pilote le flux a augmenté en présence d’US, mais une réduction des caroténoïdes et des microorganismes du JC a été observée en lien avec le mode d’application des US. Les mesures de SAXS durant la MFT du jus-modèle ont révélé pour la première fois l’accumulation de pectine à la Sm induisant une diminution du flux, aussi, ils ont mis en évidence un effet de synergie entre le débit tangentiel et l’US pour la réduction des couches concentrée
