Synthesis, characterization, structure, DNA binding and biological activity studies of new silver(I) sacchainate complexes containing some mono- and bidentate tertiary phosphine ligands

Bu çalışmada tek ve çift dişli tersiyer fosfin ligantları içeren sekiz adet yeni gümüş(I)-sakkarinat (sac) kompleksi sentezlendi. Komplekslerin yapıları elementel analiz, IR, NMR, ESI-MS, TG-DTA ve X-ışını tek kristal kırınım yöntemi ile aydınlatıldı. Bunun yanı sıra, komplekslerin Balık Spermi DNA'sına (BS-DNA) bağlanma özellikleri UV-Görünür, floresans, viskozite ve jel elektroforez teknikleri kullanılarak incelendi. Ayrıca komplekslerin, bazı bakteriler (Staphylococcus aureus, Escherichia coli ve Salmonella typhi) ile A549 ve MCF-7 gibi farklı kanser hücre soyları üzerindeki antibakteriyal ve antikanser aktiviteleri araştırıldı. Sentezde trifenilfosfin(PPh3), siklohekzildifenilfosfin (PPh2Cy), disiklohekzilfenilfosfin (PPhCy2), trisiklohekzilfosfin (PCy3), 1,1-bis(difenilfosfino)metan (dppm), 1,2-bis(difenilfosfino)etan (dppe), 1,3-bis(difenilfosfino)propan (dppp), 1,4-bis(difenilfosfino)butan (dppb) ligantları kullanıldı ve sadece dppp ligantı içeren gümüş(I)-sakkarinat kompleksinin tek kristali elde edilemedi. X-ışını tek kristal çalışmaları gümüş(I) iyonu etrafında ligantların düzenlenmesinin genel olarak doğrusal, T-şekilli ve üçgen düzlem geometride olduğunu gösterdi. Komplekslerde sac ligandı, [Ag2(sac)2(-dppm)H2O]•H2O ve [Ag(-sac)-dppb)]n'de negatif yüklü azot atomu üzerinden koordinasyon göstermektedir. [Ag(-sac)(PPh3)]2 ve {[Ag2(-sac)2(-dppe)]•H2O}n'de N ve Osülfonil, [Ag(-sac)(PPh2Cy)]2, [Ag(-sac)(PPhCy2)]2 ve [Ag(-sac)(PCy3)]n komplekslerinde ise N ve Okarbonil üzerinden iki gümüş(I) merkezi arasında köprü oluşturmaktadır. UV-Vis titrasyon ve floresans analiz çalışmaları komplekslerin BS-DNA ile interkalasyon şeklinde etkileştiğini göstermektedir. Jel elektroforez çalışmaları ise, komplekslerin pBR322 plazmid DNA ile etkileşimlerini doğrulamaktadır. Ayrıca, komplekslerin seçilen bakteri ve farklı kanser hücre soylarına karşı antibakteriyal ve antikanser aktiviteleri incelendi. Komplekslerden özellikle [Ag(-sac)(PPh3)]2'in aktivitesinin referans alınan AgNO3, AgSD (gümüşsülfadiazin) ve cisplatine göre oldukça iyi olduğu belirlendi.MSc Thesis In this study, eight new silver(I) saccharinate complexes containing mono- and bidentate tertiary phosphines were synthesized. The complexes were characterized by elemental analysis, IR, NMR, ESI-MS, TG-DTA techniques and single crystal X-ray diffraction method. In addition, the DNA binding properties of these complexes for Fish Sperm DNA (FS-DNA) were investigated using UV-Vis, fluorescence, viscosity and gel electrophoresis measurements. Furthermore, the antibacterial and anticancer activities of complexes on different bacteria (Staphylococcus aureus, Escherichia coli, Salmonella typhi), and A549 and MCF-7 cancer cell lines were studied. In synthesis triphenylphosphane (PPh3), cyclohexyldiphenylphosphane (PPh2Cy), dicyclohexylphenylphosphane (PPhCy2), tricyclohexylphosphane (PCy3), 1,1-bis(diphenylphosphino)methane (dppm), 1,2-bis(diphenylphosphino)ethane (dppe), 1,3-bis(diphenylphosphino)propane (dppp) and 1,4-bis(diphenylphosphino)butane (dppb) were used and only the cyrstal of complex of dppp was not obtained. X-ray diffraction studies have demonstrated that in general the silver(I) ion has linear, distorted T-shaped and trigonal coordination geometries. In the complexes of [Ag2(sac)2(-dppm)H2O]•H2O ve [Ag(-sac)(-dppb)]n, sac coordinates to Ag(I) through the negative charged nitrogen atom. In [Ag(-sac)(PPh3)]2 and {[Ag2(-sac)2(-dppe)]•H2O}n sac acts as a bridging ligand via N and Osulfonyl atoms, whereas in [Ag(-sac)(PPh2Cy)]2, [Ag(-sac)(PPhCy2)]2 ve [Ag(-sac)(PCy3)]n via N and Ocarbonyl atoms. UV-Vis and fluorescence analyses indicate that the complexes bind to DNA by a single intercalative mechanism. The gel electrophoresis assay further confirms their binding with the pBR322 plasmid DNA.
