The effects on taurine on oxidant and antioxidant systems in experimentally induced type 2 diabetes

Diyabetes mellitus'ta kan glukoz ve lipit düzeyinde gözlenen artış sonucuoluşan oksidatif stres, diyabetin komplikasyonlarının gelişmesinde önemli bir rolesahiptir. Taurin, kan glukoz ve lipit düzeyini düşürücü etkisi nedeniyle oksidatifstresi azaltabilir. Bu çalışmada streptozotosin-nikotinamit ile tip 2 diyabetoluşturulmuş sıçanlarda taurinin hipoglisemik, oksidan ve antioksidan sistemlerüzerine etkisi araştırıldı. Nikotinamitin (45mg/kg) intraperitonealenjeksiyonundan 15 dk sonra streptozotosin (65 mg/kg) enjeksiyonu ile tip 2diyabet oluşturuldu. Taurin (%1) içme suyuna 5 hafta süre ile eklendi. 32 adetWistar türü erkek sıçanlar rastgele kendi aralarında dört gruba ayrıldı; kontrol (K),kontrol + taurin (K + T), diyabet (D), diyabet + taurin (D + T). K + T grubundaK grubuna göre serum trigliserit, doku ve plazma malondialdehit düzeylerindeanlamlı azalma saptanırken, total antioksidan kapasitesinde anlamlı artış saptandı.D + T grubunda diyabet grubuna göre serum total kolesterol, trigliserit, kanglukoz, plazma ve doku malondialdehit düzeylerinde anlamlı azalma saptanırken,serum insülin, kan glutatyon peroksidaz, eritrosit süperoksit dismutaz,paraoksonaz ve arilesteraz aktivitesinde ise anlamlı artış olduğu saptandı.Sonuç olarak çalışmamızda taurinin, antihiperglisemik veantihiperlipidemik ve antioksidan özelliği ile tip 2 diyabette oluşan oksidatif stresekarşı koruyucu ve/veya önleyici etkisinin olduğu ve diyabette tedaviye ek olarakkullanılmasının yararlı olabileceği sonucuna varıldı.Oxidative stress which occurs as a result of increased blood glucose and lipidlevels plays an important role in the progression of the complications of diabetes. Sincetaurine is able to reduce blood glucose and lipid levels, it is thought to decreaseoxidative stress. This study was designed to investigate the effects of taurine onhypoglycemic, oxidant and antioxidant systems in streptozotocin -nicotinamide inducedtype 2 diabetes. Subjects were made type 2 diabetes by injecting nicotinamide(45mg/kg) intraperitoneally 15 min before injection of streptozotocin (65 mg/kg).Taurine (1%) was supplemented in drinking water for 5 weeks.Thirty two male Wistarrats were randomly divided into four groups; control (C), control + taurine (C + T),diabet (D), diabet + taurine (D + T). Serum trigliseride, tissue and plasmamalondialdehyde levels were observed to be significantly reduced while serum totalantioxidant capacity were significantly increased in C + T group when compared withcontrol group. Serum total cholesterol and triglyceride, blood glucose, plasma and tissuemalondialdehyde levels were signigicantly reduced while serum insulin, bloodglutathione peroxidase and erythrocyte superoxide dismutase, serum paraoxonase andarylesterase activities were significantly increased in D + T group when compared withD group.In conclusion, because of its antihyperglycemic and antihyperlipidemic andantioxidant features, taurine plays a protective and preventive role against oxidativestress in type 2 diabetes and it can be used to supplement and support the treatment ofdiabetes
