Effect of enterocins on planktonic cells and biofilm of Salmonella Typhimurium and Escherichia coli serovars

Escherichia coli and Salmonella Typhimurium are bacteria that present risks to human health, being able to form biofilms on abiotic surfaces, constituting an important source of contamination in the food industry, and an alternative for their control are the bacteriocins, because they are able to eliminate bacteria through the formation of pores in the cellular membrane. The objective of this study was to evaluate the action of bacteriocins produced by Enterococcus in planktonic cells and biofilm cells formed by Salmonella and sorovars Escherichia coli. Cell-free supernatant (CFS) containing enterocin produced by 3 Enterococcus isolates (MF2, MF5 and L3) were used in the plate antagonism test. Subsequently, the enterocin was tested in formed biofilm, being measured by the biomass measurement after application of violet crystal. The synergistic action among the enterocins was verified by the construction of a response surface by mixing planning. All Enterococcus isolates tested have more than two genes for expression of enterocin. The crude CFS containing enterocin showed inhibition against the indicator bacteria at all times of contact tested (6h and 18h). The enterocin expressed by the L3 isolate showed the best inhibition profile on planktonic cells. In the biofilm, the best time of inhibition occurred in the time 18h. The response surface for the synergism test showed that there was small synergistic binary interaction between the CFS MF2 and MF5 against one of the pathogens. The results show that the enterocins produced by these lactic acid bacteria (BAL) have potential for use as preservatives in foods, since the action spectrum also encompasses Gram negative bacteria in contrast to nisin, which is currently used.Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES)Escherichia coli e Samonella Typhimurium são bactérias que apresentam riscos à saúde humana, sendo capazes de formar biofilmes em superfícies abióticas, constituindo uma importante fonte de contaminação na indústria de alimentos. Por serem estruturas complexas, são resistentes a diversos sanitifizantes, e uma alternativa para seu controle são as bacteriocinas, pois são capazes de eliminar bactérias através de formação de poros na membrana celular. O objetivo deste estudo foi avaliar a ação de bacteriocinas produzidas por Enterococcus em células planctônicas e células de biofilme formado por Salmonella Typhimurium e Escherichia coli. Foram utilizados sobrenadante livre de células (CFS) contendo enterocina produzida por 3 isolados de Enterococcus ( MF2, MF5 e L3), no teste de antagonismo em placas. Posteriormente, a enterocina foi testada em biofilme formado, sendo mensurado pela medida da biomassa após aplicação de cristal violeta. A ação sinérgica entre as enterocinas foi verificado pela construção de uma superfície de resposta por planejamento de misturas. O CFS bruto contendo enterocina apresentou inibição contra as bactérias indicadoras, em todos os tempos de contato testados (6h e 18h). A enterocina expressada pelo isolado L3 foi o que apresentou o melhor perfil de inibição sobre células planctônicas. Já no biofilme o melhor tempo de inibição ocorreu no tempo 18h. A superfície de resposta para o teste de sinergismo mostrou que houve pequena interação binária sinérgica entre os CFS MF2 e MF5 contra um dos patógenos. Os resultados obtidos mostraram que as enterocinas produzidas por esses Enterococcus tem potencial para uso como conservantes em alimentos, visto que o espectro de ação abrange também as bactérias Gram negativas ao contrário da nisina, que é usada atualmente
