Mitokondriyal mutantların bira fermantasyonu üzerine etkisi

TEZ5824Tez (Yüksek Lisans) -- Çukurova Üniversitesi, Adana, 2006.Kaynakça (s. 46-55) var.viii, 55 s. : rnk. ; 29 cm.The objective of this study was to determine the effect of mitochondrial mutants of bottom fermentation yeast of Saccharomyces carlsbergensis NCYC 1056 and top fermentation yeast of Saccharomyces cerevisiae NCYC 1006 on beer quality. Wort inoculated with mitochondrial mutants at 1x10[7] cell/mL. Wort inoculated with main yeasts at 1x10[7] cell/mL was used as control. During the fermentation the yeast growth and fermentation rate were measured. Specific gravity, ethyl alcohol, pH, titration acidity, total and reducing sugars and aroma compounds were determined in the beers. According to the results obtained, fermentation rate and growth of yeast decreased in the samples produced by mitochondrial mutants. In addition, mitochondrial mutants produced lower amounts of ethyl alcohol and aroma compounds than the main yeasts. The amount of diacetyl which is off flavour and play an important role on the beer quality were found higher in the samples produced by mitochondrial mutant of Saccharomyces cerevisiae NCYC 1006 than that of control.Beer, Mitochondri, Mitochondrial Mutant, Mutation, Saccharomyces spp. Bu çalısmanın amacı, alt fermantasyon bira mayası Saccharomyces carlsbergensis NCYC 1056 ve üst fermantasyon bira mayası Saccharomyces cerevisiae NCYC 1006 mayaları ile yürütülen bira fermantasyonlarından izole edilen mitokondriyal mutantların bira kalitesi üzerine etkisini incelemektir. Çalısmada malt sıralarına izole edilen mitokondriyal mutantlar 1x10 [üzeri]7 hücre/mL oranında ilave edilmistir. Mitokondriyal mutant kullanılmayan dogrudan 1x10[üzeri]7 hücre/mL bira mayası ilave edilen deneme kontrol olarak kullanılmıstır. Çalısmada fermantasyon süresince maya gelisimi ve fermantasyon hızı belirlenmistir. Elde edilen biralarda yogunluk, etil alkol, titrasyon asitligi, pH, indirgen seker ve toplam seker analizleri ile aroma maddeleri tayini yapılmıstır. Mitokondriyal mutantlarının kullanıldıgı denemelerde fermantasyon hızı ve maya gelisimi kontrol örnegine göre daha yavas bulunmustur. Ayrıca, mitokondriyal mutant kullanımı ile elde edilen biralarda etil alkol ve aroma maddeleri miktarı kontrole göre daha düsük degerlerde belirlenmistir. Bira üretiminde istenmeyen bir aroma maddesi olan diasetil S. cerevisiae NCYC 1006 mayasının mitokondriyal mutantları ile üretilen biralarda daha yüksek bulunmustur.Bu çalışma Ç.Ü.
