Farklı ortam koşullarının (ışık, sıcaklık, besin eksikliği ve havalandırma) haematococcus pluvialis flotow'da büyüme ve astaksantin miktarına etkisi

TEZ7100Tez (Yüksek Lisans) -- Çukurova Üniversitesi, Adana, 2008.Kaynakça (s.36-42) var.vii, 43 s. : rnk.res. ; 29 cm.In the study carried out to determine the effects of light intensity, temperature, nitrogen deficiency and aeration on the vegetative growth and form of the cysts of the green alga Haematococcus pluvialis in the laboratory conditions, vegetative growth of the cultures were found similar at the light intensities of 27 and 48 ?mol m-2s-1 and % 2-10 inoculation amounts. H. pluvialis cultures were exposed to the 177 and 379 ?mol m-2s-1 light intensities and N-deficient condition to form cyst and accumulate astaxanthin. While 27 and 48 ?mol m-2s-1 light intensities caused the accumulation of astaxanthin at the low cell concentrations, 177 ?mol m- 2s-1 irradiance was insufficient to turn red the cells in the N-deficient medium and high cell densities. The highest astaxanthin content of % 3.60 was obtained from the cultures aerated, at the irradiance of 379 ?mol m-2s-1 and 24 °C. In the study, it was observed that 35 °C temperature and 177 ?mol m-2s-1 irradiance caused the death of the cells.Laboratuvar koşullarında kültüre alınan yeşil alg Haematococcus pluvialis vejetatif büyümesine ve kiste dönüşümüne, ışık, sıcaklık ve havalandırma etkisini belirlemek amacıyla yürütülen çalışmada 27 ve 48 ?mol m-2s-1 aydınlanma ve % 2- 10 aşılama miktarlarının vejetatif gelişmeye önemli bir etkisi olmadığı belirlenmiştir. Kist oluşumunun sağlanması için 177 ve 379 ?mol m-2s-1 aydınlanma sağlanmış ve aynı zamanda besi ortamında azot eksiltmesi yapılmıştır. Düşük hücre yoğunluğunda 27 ve 48 ?mol m-2s-1 aydınlanma şiddetleri büyümenin 11. gününde astaksantin birikimine neden olurken, yüksek hücre yoğunluğu ve azot eksik ortamda 177 ?mol m-2s-1 ışık şiddetinin kist oluşumu için yeterli olmadığı saptanmıştır. En yüksek astaksantin miktarı %3.60 ile 379 ?mol m-2s-1 aydınlanma şiddetinde, 24 ºC sıcaklıkta ve havalandırma yapılan kültürlerde saptanmıştır. Çalışmada 35 ºC sıcaklığın ve 177 ?mol m-2s-1 aydınlanmanın hücrelerde ölüme sebep olduğu gözlenmiştir.Bu çalışma Ç.Ü. Bilimsel Araştırma Projeleri Birimi Tarafından Desteklenmiştir.
