Microbiological properties of milks preserved by the activation of lactoperoxidase system and in cold storage

Araştırmada, 12:8 mg/l ve 24:16 mg/l Tiyosiyanat: Hidrojenperoksit (SCN-:H2O2) ilavesi ile laktoperoksidaz sistemi aktive edilen sütler 30°C'de 6 saat süreyle bekletilmişlerdir. Soğutularak 4±rC'de tutulan, ayrıca kontrol olarak da SCN~ H2O2 katılmamış ve 30°C'de tutulan süt örnekler! de aynı süre bekletilmişlerdir. Bekletmenin 0., 3. ve 6. saatlerinde sütün pH değeri, asillik derecesi ve bazı mikroorganizma grubu içeriklerindeki değişimler incelenmiştir. Laktoperoksidaz sisteminin aktivasyonunun, sütlerin toplam aerob mezofil ve psikrotrof bakteri gruplarının, mezofil ve termofil laktik asit bakteri "gruplarının, enterobakterlerin ve küf-mayaların gelişimini önemli düzeyde etkilediği görülmüştür (p<0,05). Soğutmanın da toplam aerob psikrotrof bakteriler dışında, incelenen diğer tüm mikroorganizma gruplarını önemli düzeyde etkilediği bulunmuştur. SCNT: H2O2 konsantrasyonunun artmasının, sistemin antimikrobiyel etkisini artırdığı belirlenmiştir. Laktoperoksidaz sisteminin aktivasyonunun soğutma kadar etkin olduğu, toplam aerob mezofil ve psikrotrof bakterilerle, enterobakterler üzerinde soğutmadan daha fazla etkili olduğu görülmüştür. Soğutma işlemi ve laktoperoksidaz sisteminin aktivasyonu, sütlerin pH ve asillik değerleri üzerinde önemli ve yaklaşık aynı düzeyde etkili bulunmuştur (p<0,05).Activation of the lactoperoxidase system during raw milk storage and its effect on the microbiological properties of milk investigated. Thus, milks were separated four parts. Milks that contained 12:8 mg/l and 24:16 mg/l SCN~: H202 were kept at 30°C for 6 hours. The third and the last one without addition of SCN~: H202 was kept at 4°C and 30°C for 6 hours, respectively. The last one used as a control. Changes of pH value, acidity and microbiological content were determined at 0., 3. and 6. hours of this period. Results Indicated that total mesophile aerobic and psychrotrophs bacteria counts, mesophile and thermophilic lactic acid bacteria counts, enterobacter and yeast/mould count were all affected by activation of lactoperoxidase system (<0.05). Cooling of milk also affected microbial content except total psychrotrophs aerobic bacteria. It was determined that increasing of SCN~: H2O2 concentration increasing antimicrobial effect of lactoperoxidase system. It was founded that activation of lactoperoxidase system as effective as cooling and it was more effective than cooling on total mesophilic aerobic, psychrotrophs bacteria and enterobacter.
