The relationship of genetic susceptibility and aggressive periodontitis (early onset periodontitis)

Agresif periodontitis (AP), erken yaşlarda şiddetli periodontal ataçman kaybı ile karakterize, hızlı ilerleyen bir grup periodontal hastalığı kapsamaktadır. Az rastlanmasına rağmen, etiyolojisi ve patogenezini anlamayı hedefleyen çok sayıda araştırma yapılmıştır. AP oluşumundaki genetik riski belirlemeye yönelik olarak yapılmış, çeşitli aile ve populasyon çalışmaları mevcuttur. AP'de gözlenen ailesel kalıtımı destekleyen genetik geçişi belirlemek amaçlı segregasyon ve linkage analizleri yapılmıştır. Günümüzde AP'de etkili olan modifiye edici hastalık genleri ile ilgili çok az bilgi mevcuttur. Sitokinler gibi immün medyatörleri kodlayan genlerdeki varyasyonlar, AP ile ilişkili olabilecek yatkınlık faktörleri için kullanılabilecek potansiyel hedef lerdir.Bu derlemede AP'in genetik yatkınlık ile ilişkisi hakkındaki son gelişmeler ve genetikle ilgili yorumlarda sıkça karşılaşacağımız çeşitli terimler özetlenmeye çalışılmıştır.Aggressive periodontitis (AP) comprises a group of rapidly progressive forms of periodontitis, characterized by severe destruction of periodontal attachment apparatus at an early age . In spite of its rare occurrence, AP has been the focus of many investigatons aimed at understanding its etiology and pathogenesis. The genetic risk of developing AP has been investigated by studying families and populations. To determine whether the observed familial aggregation for AP supports genetic transmission of the disease; segregation and linkage analyses had been performed. Currently very little is known about which genes may be involved in aggressive periodontitis as modifying disease genes. Genetic variation in the genes of immune mediators, such as cytokines, has been used as a potential target for susceptibility factors in relation to AP. The present review will focus on recent findings relating genetics to AP and will summarize various terms frequently used in discussions involving genetics
