Domateste, dayanıklı ve duyarlı genotiplerin melezlenmesinden elde edilen saf hatlarda demir (fe) eksikliğine tolerans

TEZ7107Tez (Yüksek Lisans) -- Çukurova Üniversitesi, Adana, 2008.Kaynakça (s.45-49) var.x, 50 s. : rnk.res. ; 29 cm.The first aim of this study was the determination of iron (Fe) deficiency tolerance levels of the reciprocal recombinant inbred lines (RILs) in tomato. The RILs (F6 generation) have been created using tolerant and sensitive inbred parents for long time studies since 1994. The second aim was the obtaining data which can be used to map the quantative trait loci (QTL) for iron (Fe) deficiency tolerance in tomato. In this study, totally 224 genotypes have been used. 107 genotypes were contained the cytoplasma of the resistant parent-Roza and 117 genotypes were contained the cytoplasma of the sensitive parent-227/1. The plants were grown in growth chamber in hydroponic medium. Thirty-five days old plants were subjected to Fe deficiency stress by 10 mM NaHCO3. The experiment repeated 5 times and each experiment was continued for 57 days. At the end of the experiments, leaf chlorosis, active and total Fe concentrations have been determined. The measured parameters of the RILs have been shown the normal distributions.Bu tez çalışmasında, 1994 yılından beri yürütülen çalışmalarla oluşturulan domateste resiprokal rekombinant saf hatların demir noksanlığı klorozuna tepkilerinin belirlenmesi ve böylece demir noksanlığı klorozuna tolerans sağlayan kantitatif lokusların (QTL) haritalanmasına katkıda bulunacak verilerin elde edilmesi hedeflenmiştir. A hattının (Roza) stoplazmasını taşıyan için 107, B hattının (227/1) stoplazmasını taşıyan için 117 olmak üzere, toplam 224 genotip kullanılmıştır. Bitkiler iklim kontrollü bitki yetiştirme odasında, su kültürü tekniği ile yetiştirilmiştir. Genç domates bitkileri 35 günlük iken 10 mM NaHCO3 ile Fe eksikliği stresine sokulmaya başlamıştır ve deneme bitkiler 57 günlük olunca tamamlanmıştır. Denemeler 5 kez tekrarlanmıştır. Her denemenin son aşamasında SPAD-metre ile klorofil seviyesi ölçülmüş ve ardından her genotipin genç yapraklarında toplam ve aktif Fe konsantrasyonu belirlenmiştir. Elde edilen sonuçlar, domateste demir eksikliğine toleransın çok genle idare edilen bir özellik olduğunu desteklemektedir.Bu çalışma Ç.Ü.
