Tyymol ve carvacrol'un in vivo genotoksik etkilerinin araştırılması

TEZ6287Tez (Doktora) -- Çukurova Üniversitesi, Adana, 2007.Kaynakça (s.107-121) var.xviii, 122 s. : res. ; 29 cm.Bu çalışmanın amacı antihelmintik, antiseptik, antifungal, antibakteriyel, antiparazitik ve antiviral olarak kullanılan carvacrol ve thymol'ün sıçan kemik iliği hücrelerinde in vivo genotoksik etkilerini araştırmaktır. Bu çalışmada carvacrol (10, 30, 50 ve 70 mg/kg'lık vücut ağırlığı) ve thymol (40, 60, 80 ve 100 mg/kg'lık vücut ağırlığı), intraperitonal uygulamada yapısal kromozom anormalliği (KA) ve total KA sayısını genel olarak tüm muamele sürelerinde (6, 12, 24 saat) ve konsantrasyonlarda kontrole göre önemli ölçüde artırmışlardır. Özellikle yüksek dozlarda hem carvacrol (70 mg/kg) hem de thymol (100 mg/kg) KA sayısını pozitif kontrol olan ethyl carbamate (EC) kadar artırmışlardır. Carvacrol sayısal KA yüzdesini en yüksek iki dozda (50 ve 70 mg/kg) kontrole göre önemli derecede artırırken, aynı şekilde thymol de yine en yüksek iki dozda (80 ve 100 mg/kg) sayısal KA yüzdesini artırmıştır. KA sayısındaki artışın doza bağlı olduğu saptanmıştır. Carvacrol ve thymol tüm dozlarda ve muamele sürelerinde MI'i kontrole göre önemli derecede düşürmüşlerdir. Carvacrol 6 ve 12 saatlik muamele sürelerinde sadece 70 mg/kg'lık dozda, 24 saatlik muamele sürelerinde ise tüm dozlarda MI'i EC kadar düşürmüştür. Thymol ise 6 saatlik muamele süresinde en yüksek iki dozda, 24 saatlik muamele süresinde de tüm dozlarda MI'i EC kadar düşürmüştür. MI'teki düşüşün de doza bağlı olduğu görülmüştür.The aim of this study was to investigate the in vivo genotoxic effects of carvacrol and thymol in bone marrow cells of rats that were used as antihelmintic, antiseptic, antifungal, antibacterial, antiparasitic and antiviral substances. In the present study, both carvacrol (10, 30, 50 and 70 mg/kg b.w.) and thymol (40, 60, 80 ve 100 mg/kg b.w.) significantly induced the structural and total chromosome abnormalities (CA) when compared with control in bone marrow cells of rats intraperitonally administered. Both carvacrol and thymol showed the similar effects with the positive control ethyl carbamate (EC) on induction of the percentage of structural and total CA at the highest concentrations (70 mg/kg b.w. and 100 mg/kg b.w., respectively). In addition, carvacrol and thymol induced the numerical CA at the highest two concentrations (50 and 70 mg/kg b.w. for carvacrol; 80 and 100 mg/kg b.w.for thymol). It was shown a dose-dependent effects on induction of structural, numerical and total CA for both carvacrol and thymol. Carvacrol and thymol decreased the mitotic index (MI) in all the concentrations and treatment times when compared with control. Carvacrol showed the similar effects with the positive control ethyl carbamate (EC) on decreasing the MI at 70 mg/kg b.w. for 6 and 12 hours and at all concentration for 24 hours treatment periods. Thymol also showed a similar effect with EC on decreasing the MI at 80 and 100 mg/kg b.w. for 6 hours and at all concentrations for 24 hours treatment periods.
