Determination of drought resistance of some american vine rootstocks using polyethylene glycol in in vitro [Bazı amerikan asma anaçlarının kurağa dayanımının in vitro’da polietilen glikol kullanılarak belirlenmesi]

In the study, it was aimed to determine the appropriate dose of Polyethylene Glycol (PEG) for early determination of drought resistance in vitro by using Kober 5 BB (drought sensitive), 110 R (drought resistant) and 1103 P (drought moderately resistant) rootstocks. For this purpose, immature nodes from summer shoots were cultured in vitro in Murashige and Skoog (MS) medium containing 1 mg L-1 BA. The shoots obtained from these nodes were transferred to MS medium containing 1 mg/L IBA with the addition of 0.0%, 2.5%, 5.0%, 7.5% and 10.0% PEG in order to create drought stress in vitro. After six weeks of culture, the level of damage on shoots, plant height, number of nodes, average length and number of roots, plant wet and dry weight, root wet and dry weight, chlorophyll and plant nutrient contents were examined. As a result, With increasing doses of PEG, it was determined that the degree of damage increased, growth and development decreased, plant fresh and dry weight and root fresh and dry weight decreased. Plant height was measured as 6.5 cm in the control application while it was measured 1.7 cm in 7.5% and 10% PEG. Plant wet and dry weight (0.257 and 0.042 g, respectively) and root wet and dry weights (0.218 and 0.023 g, respectively) of control plants were higher than PEG treatments. SPAD readings were also found to decrease inversely with increasing PEG doses. Considering the plant element contents, P, K, Ca and Mn and Cu element values were found to be lower in different PEG applications than control plants. In conclusion, it was concluded that 2.5% and 5.0% doses of PEG with shoot and root parameters could be used in vine rootstocks in order to determine drought resistance early in in vitro conditions. © 2019, Centenary University.
