In vitro genotoxic effets of benzoyl peroxide in human peripheral lymphocytes.

TEZ5507Tez (Yüksek Lisans) -- Çukurova Üniversitesi, Adana, 2005.Kaynakça (s. 108-131) var.xiii, 132 s. : rnk.res. ; 29 cm.The aim of this study was to investigate the possible genotoxic effects of benzoyl peroxide, which used as bleaching agent in flour, by determining in vitro sister chromatid exchange (SCE), chromosome aberration (CA) and micronucleus (MN) tests in human peripheral lymphocytes. The cells were treated with 25, 50, 75 and 100 ug/ml benzoyl peroxide for 24 h and 48 h treatment periods. Benzoyl peroxide generally induced SCEs, but this effect was found significant only at the highest concentration (100 ug/ml) for 48 h treatment period in the present study. Also benzoyl peroxide increased both percentage of abnormal cells and number of chromosome aberrations per cell (CAs/Cell) at 75 ug/ml and 100 ug/ml concentrations for 24 h treatment period and at all concentrations for 48 h treatment period in human peripheral lymphocytes. This increase was dosedependently manner. In addition to this, benzoyl peroxide affected negatively both replication index (RI) and mitotic index (MI). However this effect was found statistically significant only for 48 h treatment period. Benzoyl peroxide generally induced micronucleus formation. Besides, this effect was statistically significant at 75 ug/ml and 100 ug/ml concentration for 48 h treatment period when compared with control.Bu çalışmanın amacı, unda beyazlatma maddesi olarak kullanılan benzol peroksit'in genotoksik etkisinin olup olmadığını insan periferal lenfositlerinde in vitro kardeş kromatid değişimi (KKD), kromozom aberasyonu (KA) ve mikronükleus (MN) testleri ile araştırmaktır. Hücreler 25, 50, 75 ve 100 ug/ml konsantrasyonlarındaki benzol peroksit ile 24 ve 48 saat muamele edilmiştir. Bu çalışmada, benzol peroksit'in genel olarak KKD'yi arttırdığı, ancak bu artışın sadece en yüksek konsantrasyondaki (100 ug/ml) benzol peroksit ile 48 saatlik muamele süresinde istatistiksel olarak önemli olduğu saptanmıştır. Ayrıca benzol peroksit insan periferal lenfositlerinde 24 saatlik muamele süresinde 75 ig/ml ve 100 ug/ml konsantrasyonlarda, 48 saatlik muamele süresinde tüm konsantrasyonlarda anormal hücre yüzdesini ve hücre başına düşen kromozom aberasyonu sayısını (KA/Hücre) arttırmıştır. Bu artış doza bağlı bir durum göstermiştir. Bunun yanında benzol peroksit'in 24 ve 48 saatlik muamele sürelerinde genel olarak hem replikasyon indeksini (RI) hem de mitotik indeksi (MI) olumsuz yönde etkilediği,ancak bu durumun sadece 48 saatlik muamele süresinde istatistiksel olarak önemli olduğu belirlenmiştir. Ayrıca benzol peroksit'in insan periferal lenfositlerinde MN oluşumunu uyardığı, ancak bu artışın, yüksek iki konsantrasyonda (75 ug/ml ve 100 ug/ml) 48 saatlik muamele süresinde kontrole göre önemli olduğu saptanmıştır.Bu çalışma Ç.Ü.
