A method of DNA extraction is given which is suitable for use with birch (Betula spp.) material collected from natural populations. Such material is frequently old and insect-damaged, and contains high levels of polyphenols. The method relies on repeated extraction of the material with a high molarity urea phosphate buffer, and yields DNA suitable for RFLP analysis
