Investigation of K-ras and BRAF gene mutations by molecular genetic methods in colorectal cancer patients

Güncel bulgular KRK'li hastalarda K-ras ve BRAF gen değişikliklerinin potansiyel prognostik ve prediktif değerlerini göstermektedir. Bizim çalışmamızın amacı kolorektal kanserli hasta gurubunda K-ras ve BRAF genlerindeki mutasyonları tanımlamak ve çeşitli klinopatolojik parametrelerle ilişkisini değerlendirmektir.Bu çalışmada, kolorektal kanser ve onların normal çevre dokularının alındığı 50 hastanın parafine gömülü doku blokları kullanılarak K-ras onkogeni kodon 12, 13, 61 ve BRAF onkogeni kodon 600 nokta mutasyonları ile tümörün klinikopatolojik karakteristiği araştırılmıştır. BRAF geni ve K-ras genindeki mutasyonlar doğrudan dizi analizi ile incelendi. İstatistiksel analizde p< 0.05 değeri anlamlı kabul edildi.50 KRK'li hasta içinde %30 (n=15) K-ras mutasyonu saptadık. Kodon 12, kodon 13 ve kodon 61'deki mutasyon sıklıkları sırasıyla %22 (11/50), %6 (3/50) ve %2 (1/50) idi. BRAF mutasyonları nadirdi ve sadece bir hasta kodon 600 mutasyon taşıyıcısıydı.Sonuçlarımız tümör lokalizasyonunun K-ras gen mutasyonu ile anlamlı ilişki gösteren tek faktör olduğunu gösterdi. Yaş, cinsiyet, tümör diferansiyasyonu, tümör morfolojisi ve evre gibi diğer klinopatolojik özellikler K-ras gen mutasyonları ile pozitif ilişki göstermedi.Recent evidence highlights the potential prognostic and predictive value of BRAF and K-RAS gene alterations in patients with colorectal cancer.
