Researches on the in vitro androgenesis and obtaining haploid plants in some eggplant genotypes

Patlıcanda haploid bitki eldesinde anter kültürünün kullanılabilirliğini belirlemek amacıyla yapılan bu çalışmada, Solanum melongena L. türüne ait Pala, Kemer, Topan, Aydın Siyahı, Adana ve Manisa yerli çeşitleri; Urfa Yerlisi populasyonu; Munica, Baluroi, Mileda, Ancha, Leila, Barbentane, Bellissima ve Purpurea Fx hibrid çeşitlerinden oluşan toplam 15 patlıcan genotipi kullanılmıştır. 1994 yılında yapılan ön denemelerde, hiçbir genotipte haploid embriyo veya bitki elde etmek mümkün olmamış, yalnızca değişen oranlarda kallus oluşumu gözlenmiştir. Genel kallus oluşum oranı %30.00 olmuştur. 1995 yılındaki denemelerde, yine haploid embriyo veya bitki elde edilememiş yalnızca kallus elde edilebilmiştir. Elde edilen kallus oranı Pala, Kemer, Topan ve Aydın Siyahı çeşitlerinde sırasıyla %15.12, 20.0, 24.0 ve 26.42 olmuştur. 1996 yılında yapılan denemelerde ise Kemer çeşidinde C ortamında %3,67, MS ortamında %2.05 oranında, Urfa Yerlisi çeşidinde ise C ortamında %4.91, MS ortamında %1.84 oranında haploid embriyo elde edilmiştir. 1998 yılında yapılan denemelerde de, anterlerden embriyoid elde etmek mümkün olmuş, embriyoid oluşum oranlan, Adana çeşidinde MS ortamında %1.58; Barbentane çeşidinde C ortamında %2.72, MS ortamında %2.63; Leila çeşidinde C ortamında %2.43 şeklinde gerçekleşmiştir. Patlıcanda, anterden haploid bitki eldesi için 5 mg/1 2,4-D ve 5 mg/1 kinetin ilave edilmiş olan C ortamı ile, 4 mg/1 NAA ve 1 mg/1 kinetin ilave edilmiş olan MS ortamı uygun ortamlar olarak bulunmuştur.This study was carried out with the aim of determining the usability of anther culture in obtaining haploid plants in eggplant (Solatium melongena L.), using the domestic cultivars Pala, Kemer, Topan, Aydın Siyahı, Adana and Manisa the domestic population Urfa Yerlisi; and the Fi hybrid cultivars Munica, Baluroi, Mileda, Ancha, Leila, Barbentane, Bellisima and Purpurea. In the preliminary trials in 1994, haploid embryos and plants could not be obtained in anyone of the genotypes, but only calli at different rates were obtained. The overall callus formation rate was 30.00%. In the trials in 1995, no haploid plants or embryos were obtained again, but only calli developed. The callus formation rates were 15.12, 20.0, 24.0 and 26.42% in cvs. Pala, Kemer, Topan and Aydın Siyahı, respectively. The obtention of haploid embryos and plantlets was achieved in the trials in 1996. Haploid embryos were obtained at the rates of 3.67% and 2.05% in C and MS media respectively in ev. Kemer; and at the rates of 4.91% and 1.84%, respectively in ev. Urfa Yerlisi. Embryos were also obtained in the trials carried out in 1998. The embryoid formation rates were 1.58% in MS medium in ev. Adana; 2.72% and 2.63% in C and MS media respectively in cv. Barbentane; and 2.43% in C medium in cv. Leila.
