Determination of phenolic compounds by liquid chromatography and mass spectrometry and antioxidant activity of some of raspberry (Rubus idaeus L.) and blackberry (Rubus fruticosus L.) cultivars grown in Bursa

Bursa da yetiştirilen beş ahududu (Aksu kırmızısı, Rubin, Newburgh, Hollanda boduru, Heritage) ve dört böğürtlen (Bursa 1, Bursa 2, Chester, Jumbo) çeşidinin fenolik madde içerikleri taze, ilk dondurulmuş ve altı ay süreyle -220C de muhafaza edildikten sonra LC-MS/MS ile tayin edilmiştir. Ellagik asit ahududu ve böğürtlen türlerinin temel fenolik maddesi olduğu tesbit edilmiştir. Ellagik asit (1350,36-727,90 mg/kg taze meyve), feruik asit (820,78-338,27 mg/kg taze meyve), kafeik asit (754,85-202,78 mg/kg taze meyve), para-kumarik asit (361,68-142,63 mg/kg taze meyve), para-hidroksibenzoik asit (534,20-233,29 mg/kg taze meyve) and kuersetin (46,97-27,31 mg/kg taze meyve) ahududu çeşitlerinin, ellagik asit (1828,07-1555,13 mg/kg taze meyve), ferulik asit (757,69-413,82 mg/kg taze meyve), kafeik asit (736,85-337,89 mg/kg taze meyve), para-kumarik asit (877,45-287,15 mg/kg taze meyve) ve kuersetin (74,69-56,78 mg/kg taze meyve) böğürtlen çeşitlerinin su ekstraklarında tayin edilmiştir. Ahududu çeşitlerinin fenolik bileşik içerikleri farklılıkları böğürtlen çeşitlerinden daha fazladır. Altı ay süreyle -220C de muhaza edilen ahududu ve böğürtlen çeşitlerinin fenolik bileşik içeriklerinin başlangıç miktarlarına göre ortalama iki kat azaldığı belirlenmiştir.İkinci olarak ahududu ve böğürtlen türlerinin toplam fenol, toplam flavonoid, toplam antosiyanin, askorbik asit miktarları ile ABTS, DPPH ve CUPRAC metotları ile antioksidan aktivite değerleri UV-VIS spektrofotometresi ile tayin edilmiştir. Ahududu ve böğürtlen çeşitleri ABTS, DPPH, CUPRAC, toplam fenol ve toplam flavonoid içerikleri metanol ekstraklarında, toplam antosiyanin ve askorbik asit içerikleri ise su ekstraklarında yüksek değerlere sahiptir. Ahududu ve böğürtlen çeşitlerinin antioksidan aktivitelerinin çeşitlerin fenolik içerikleriyle direk olarak ilişkili olduğu bulunmuştur. Antioksidan aktivite ile toplam fenol, toplam flavonoid ya da askorbik asit arasında doğrusal ilişki olduğu gözlenmiştir. Elde edilen sonuçlar ahududu ve böğürtlen türlerinin zengin antioksidan kaynağı olduğunu göstermektedir.The phenolic compounds in five raspberry (Aksu kırmızısı, Rubin, Newburgh, Holland boduru, Heritage) and four blackberry (Bursa 1, Bursa 2, Jumbo, Chester) cultivars grown in Turkey were determined by LC-MS/MS in fresh, just frozen and stored fruits at -22 0C for 6 months period. Ellagic acid was determined as main phenolic compounds in raspberry and blackberry cultivars. The major phenolic compounds in water extracted samples were ellagic acid (1350,36-727,90 mg/kg fresh fruit), ferulic acid (820,78-338,27 mg/kg fresh fruit), caffeic acid (754,85-202,78 mg/kg fresh fruit), p-coumaric acid (361,68-142,63 mg/kg fresh fruit), p-hydroxybenzoic acid (534,20-233,29 mg/kg fresh fruit) and quercetin (46,97-27,31 mg/kg fresh fruit) in raspberry and ellagic acid (1828,07-1555,13 mg/kg fresh fruit), ferulic acid (757,69-413,82 mg/kg fresh fruit), caffeic acid (736,85-337,89 mg/kg fresh fruit), p-coumaric acid (877,45-287,15 mg/kg fresh fruit) and quercetin (74,69-56,78 mg/kg fresh fruit) in blackberry. The varietal differences in the phenolic compound contents were larger among the raspberry cultivars than the blackberry cultivars. After storage of raspberry and blackberry at -22 0C for 6 months, the amount of individual phenolic compounds decreased by two times on average.Raspberry and blackberry cultivars were also assayed for antioxidant activity determined as ABTS, DPPH. and CUPRAC, total phenol, total flavonoid, total anthocyanin and ascorbic acid contents. Raspberry and blackberry had the highest ABTS, DPPH, CUPRAC, total phenol and total flavonoid contents in methanol extracts, whereas total anthocyanin and ascorbic acid contents were high in water extracts. The antioxidant activity of the raspberry and blackberry was directly related to the total amount of phenolics found in the raspberry and blackberry. A linear relationship was observed between antioxidant activities and total phenols, total flavonoids or total anthocyanins.
