dissertationPoliovirus RNA synthesis in vitro was studied in an attempt to determine the conditions required for accurate product synthesis. Analysis of in vitro reaction products by gel electrophoresis, sucrose gradient sedimentation, hybridization and RNase T(,1) oligonucleotide fingerprints revealed that crude cytoplasmic extracts synthesize products which are identical to those made in infected cells; whereas, detergent-extracted complexes fail to synthesize full-length single-stranded RNA. Both complexes synthesize predominantly plus-stranded RNA, and detergent treatment inhibits synthesis of full-length single-stranded RNA by complexes which, untreated, are capable of making this product.
