Determination of Effect a-Lipoic Acid on DNA Damage, Protein Oxidation, Lipid Peroxidation and Some Biochemical Paramets at Subchronic Thinner Inhalated Rats.

The misusage of volatile Chemicals for pleasure rather than industrial purposes has been increasing for a long time. Some of the well known side effects of thinner and thinner like compounds are: causing the increase of ROS, damage to cellular components e.g. DNA, proteins, lipids and cellular aging. Living organisms defend and tolerate themselves against to their harms. The present study was carried out to determine both the effects of sub-chronic thinner inhalation on oxidant-antioxidant balance, and relation between toxicity and oxidative stress and possible protective effect of a-lipoic acid against to thinner toxication in rats. In the study, rats, Sprague-Dawley, were divided into 5 groups as follow: Control (K), Olive oil (Z), a-lipoic acid (LA), Thinner (T), and a-lipoic acid+thinner (LAT). Each group was composed of 15 rats and study lasted 8 weeks. After completing the animal studies, MDA, GSH, MetHb, Toluol, and mononuclear leucocyte damage levels, protein oxidation, NOx metabolites, TAC, Glucose, Total Cholesterol, triglyceride, HDL-Cholesterol, LDL-Cholesterol, AST and ALT levels were determined in the blood specimen of the rats employed. The data obtained from the study were analysed statistically by using SPSS package software and both Anova and Duncan tests were performed. p<0.05 level was accepted as statistically significant level. The results indicated that a-Lipoic acid has protective and balancing effect against to complications occured by thinner inhalation in rats.Uçucu maddelerin endüstriyel amaçlar dışında keyif verici ve uyarıcı olarak kötüye kullanımı önemli boyutlardadır. Endüstriyel amaçla yaygın kullanılan tiner ve benzeri çözücülerin inhalasyonu ROS’inin aşırı üretilmesine neden olduğu ve DNA, proteinler ve lipidlere etki ederek hücre hasarına ve yaşlanmaya neden olduğu bilinmektedir. Organizma ise serbest radikallerin zararlı etkilerini enzimatik ve enzimatik olmayan antioksidanların aktivasyonu ile dengeleyip uzaklaştırmaktadır. Çalışmamız, subkronik tiner inhalasyonunun oksidan- antioksidan denge üzerine etkilerini ve toksisitenin oksidatif stresle ilişkili olup olmadığını belirlemek ve a-lipoik asitin subkronik tiner toksikasyonuna karşı koruyucu etkilerini araştırmak amacıyla gerçekleştirilmiştir. Çalışmada, kontrol grubu (K), zeytin yağ (Z), a- lipoik asit (LA), tiner (T) ve tiner + a-lipoik asit (LAT) olmak üzere, her grupta 15 Sprague- Dawley cinsi rat içeren 5 grup oluşturulmuştur. Deneme süresi olan 8 hafta sonra deneklerden alınan kan örneklerinde; MDA, GSH, MetHb, Toluol ve Mononuklear Lökosit DNA hasar düzeyleri, protein oksidasyonu, NOX metabolitleri, TAK, Glikoz, Total Kolestrol, Trigliserid, HDL Kolestrol, LDL Kolesterol, AST ve ALT düzeyleri tespit edildi.
