Importance of the detection of telomerase activity in the patients with chronic lymphocytic and acute myeloblastic leukemia; Our first results

Ökaryotik lineer kromozomların uçlarında yer alan ve tekrarlayan DNA dizileri ile özel proteinlerden oluşan telomerler, kromozom uçlarını yıkıma karşı korurlar. Telomer boyu değişimi, telomer proteinleri ve telomeraz enzim kompleksi tarafından kontrol edilir. Telomeraz aktivitesi, fetal ve ergin testisler, fetal ovaryum, tamir gören dokuların çoğalan hücreleri, hematopoietik kök hücreleri, lenfositler, saç folikülleri ve barsak kripta hücrelerinde tespit edilmiştir. Bununla birlikte en sık olarak malignansilerin %85’inde (örneğin; hematolojik malignansiler) yüksek telomeraz aktivitesi belirlenmiştir. Bu nedenle telomeraz aktivitesinin, kanser teşhisinde önemli bir gösterge olabileceği düşünülmektedir. Çalışmamızda lösemik hücrelerde ve normal lenfositlerde telomeraz aktivitesinin tayini ve kıyaslanması amaçlanmıştır. Bu nedenle, sırasıyla Rai ve FAB kriterlerine göre yeni tanı 7 kronik lenfositik lösemi (KLL), 7 akut myeloblastik lösemi (AML) hastası ve 7 sağlıklı kişiden elde edilen toplam 21 periferik kan örneğinde telomeraz aktivitesi, TeloTAAGGG Telomerase PCR ELISAPLUS (Roche) kiti kullanılarak tayin edildi. PCR reaksiyonunun doğruluğundan emin olmak için PCR ürünleri poliakrilamid jelde yürü¬tüldü ve gümüş boyama yöntemi ile bantlar görünür hale ge¬tirildi. KLL’li hastalarda kontrol grubuna göre düşük, AML’li hastalarda ise kontrol grubuna göre yüksek oranda telomeraz aktivitesi belirlendi. Ancak gruplar arasındaki fark, istatistiksel olarak anlamlı değildi. Sonuç olarak; lösemilerin prognozu, evrelendirilmesi, hastalığın remisyonu ve tedaviye verdiği yanıtın takibinde telomeraz aktivite tayininin güvenilir bir gösterge olabileceği düşünüldü.Telomeres, which are located at the ends of eukaryotic linear chromosomes and are comprised of repetitive DNA sequences with special proteins, protect ends of chromosomes against degradation. Telomere lenght is controlled by telomere proteins and telomerase enzyme complex. Telomerase activity is not detectable in somatic cells except for fetal and adult testes, fetal ovary, proliferative cells from renewal tissues, hematopoietic stem cells, lymphocytes, hair follicles and cryptic cells in the intestine. However high telomerase activity had been detected in %85 of human malignancies (e.g. hematologic malignancies). Thus it has thought that telomerase activity is a important marker in the cancer diagnosis. In our study it had been aimed determination of telomerase activity and comparison in leukemic cells and lymphocytes. Thus telomerase activity was determined by using TeloTAAGGG Telomerase PCR ELISAPLUS (Roche) kit in total 21 peripheral blood samples collected from newly diagnosed patients according to Rai and FAB criteria respectively 7 chronic lymphocytic leukemia (CLL), 7 acute myeloblastic leukemia (AML) patients and 7 healthy persons. To ensure the truth of PCR reaction PCR products were electrophoresed on a polyacrylamide gel and bands were made visible by the method of silver staining. Telomerase activity was low in the CLL patients and high in the AML patients according to control group. But the differences between the groups statistically was not significant. As a result, it was thought that determination of telomerase activity could be significant marker for prognose, remission and therapy of leukemia
