Incorporation of non-natural amino acids into the lantibiotic lichenicidin and generation of protein hybrids of the proteins PabB and TrpE of B. subtilis

Antibiotika sind für die Therapie von Infektionskrankheiten essentiell. Die Ausbildung von Resistenzen gegen Antibiotika stellt eine der großen Herausforderungen für die medizinische Forschung zur Findung und Testierung neuer Wirkstoffkandidaten dar. Als ribosomal synthetisierte Peptidantibiotika beinhaltet die Klasse der Lantibiotika interessante Substanzen. Das gemeinsame Strukturmotiv der Lantipeptide, denen die Lantibiotika angehören, sind intramolekulare Thioetherbrücken, die posttranslational durch spezialisierte Enzyme gebildet werden. Das Klasse II-Lantibiotikum Lichenicidin wurde aus dem Gram-positiven Bakterium Bacillus licheniformis I89 isoliert und besteht aus den Untereinheiten BliA und BliB. Die Gene der Lichenicidin-Biosynthese sind in Bacillus licheniformis I89 in einem Gencluster organisiert, das erfolgreich in den heterologen Wirt Escherichia coli transferiert werden konnte. Im ersten Teil dieser Arbeit wurde die Biosynthese der Lichenicidin-Untereinheiten gezielt mit nicht-kanonischen Aminosäure-Analogen (ncAA) der Aminosäuren Methionin, Tyrosin, Lysin und Arginin modifiziert. Dabei wurde die Promiskuität des Translationssystems unter selektivem Druck in auxotrophen Escherichia coli-Stämmen zur Akzeptanz der ncAA ausgenutzt. Nachfolgend wurden Mutanten erzeugt, bei denen ausgewählte Aminosäure-Positionen durch Methionin oder Alanin ersetzt wurden. Diese Methionin-Mutanten wurden ebenfalls mit Hilfe von ncAA unter Selektionsdruck modifiziert. Die erhaltenen Mutanten und Kongenere wurden im nächsten Schritt auf ihre antibiotische Wirkung gegen den Indikatorstamm Micrococcus luteus untersucht. Die postbiosynthetische Modifikation mit Hilfe der Klick-Chemie wurde anhand der Amzidohomoalanin- und Homopropargylglycin-Kongenere der Lichenicidin- Untereinheit BliA untersucht. Die Synthese von para-Aminobenzoat und Anthranilat und deren nachgelagerten Biosynthesen des Folats und der Aminosäure Tryptophan stellen wichtige Biosynthesewege in der bakteriellen Zelle dar. Ebenso sind diese Biosynthesewege Angriffspunkte für verschiedene Antibiotika. Innerhalb dieser Biosynthesewege bilden die para-Aminobenzoat-Synthase und Anthranilat-Synthase ausgehend vom selben Edukt, Chorismat regioisomere Verbindungen. Im zweiten Teil dieser Arbeit wurden die Enzyme PabB und TrpE untersucht und ein Konzept erarbeitet, bei dem die nicht-konservierten Bereiche der Enzyme aus B. subtilis gezielt gegen die entsprechenden Bereiche des jeweils anderen Enzyms ausgetauscht werden können. Die so erhaltenen Mutanten wurden in E. coli synthetisiert und das erhaltene Produkt der enzymatischen Umsetzung massenspektrometrisch untersucht.Antibiotics are essential for the treatment of bacterial infections. Nevertheless, the formation of resistance against one or several antibiotics is a challenging factor for medical research in respect to find and test next generation antibiotics. Ribosomally synthesized peptide antibiotic are an important group of secondary metabolites. Among them, lantibiotics constitute interesting compounds such as the class II lantibiotic lichenicidin. Lichenicidin was isolated from Gram-positive bacteria Bacillus licheniformis I89. Lichenicidin is formed by two subunits BliA and BliB. Subsequent to ribosomal peptide synthesis, lantipeptides, which lantibiotics are part of, are enzymatically and posttranslationally modifed to form intramolecular thioether bridges. In Bacillus licheniformis I89, the genes for lichenicidin biosynthesis are clustered. This cluster was successfully transferred to heterologous host Escherichia coli for peptide synthesis. In the first part of this work, lichenicidin was used to introduce noncanonical amino acids (ncAA) by selective pressure induced incorporation in auxotrophic Escherichia coli strains. Methionine, lysine, arginine and tyrosine were chosen to introduce ncAAs, using the similarity of ncAA analogues to corresponding canonical amino acid to be accepted by biosynthesis machinery and posttranslational modification enzymes of lichenicidin biosynthesis. In second step, peptide mutants were generated, where elected amino acid positions were exchanged by methionine or alanine. The methionine-mutants were modified once more in vivo by using ncAAs. Subsequently, these congeners and mutants were tested against the bacteria Micrococcus luteus in inhibition assay assessing the antibacterial effects. The postbiosynthetical modification was investigated by click-reaction. Therefore, the Azidohomoalanine and Homopropargyle congeners of BliA were chosen. Synthesis of para-aminobenzoic acid and anthranilate and following pathways towards folate and tryptophan are important for bacterial cell growth. Several antibiotics are interacting with these biosynthetic pathways. Within this biosynthetic pathways, the para-aminobenzoic acid-synthase and anthranilate-synthase are producing region isomeric compounds out of the same precursor Chorismate. In the second part of this work, the enzymes PabB and TrpE where analyzed and a way was established to exchange non-conserved regions of these two enzymes from Bacillus subtilis with the corresponding region in the second enzyme.
