Evasion of apoptosis is one of the main hallmarks of cancer. The BCL-2 family of proteins are the main mediators of the intrinsic pathway of apoptosis and the interactions between the pro- and anti-apoptotic members of this family of proteins control the fate of a cell towards life or death. Anti-apoptotic members of the BCL-2 family, such as BCL-2, BCL-XL and MCL-1, are frequently overexpressed in a plethora of cancers promoting abnormal cell survival. During the last ten years, several small molecule inhibitors called BH3 mimetics were developed to inhibit these anti-apoptotic proteins.
