Lipase (EC 3.1.1.3) is one of class hydrolase enzymes that can hydrolyze triglycerides into fatty acids and glycerol. This research determine the optimum conditions of lipase production from Bacillus isolates sp.TM_2a.2 which isolated from hot spring Tamalantik, Mamasa, West Celebes with Various of substrat concentration (olive oil) is 0.2%, 0.4%, 0, 6%, 0.8% and 1%. The activity of lipase production was tested by titrimetric method. The results show that the lipase produced optimum at substrat concentration of 0,8% for 36 hours, with activity of 0,3230 U/mL and specific activity of 0,8296 U/mg. Optimum condition lipase reached at pH 7 and temperature at 40 oC with activity of 0,2584 U/mg
