RT is a key enzyme in the life cycle of HIV, and is targeted by multiple antiviral drugs. Although for most of its function RT binds a dsDNA or RNA–DNA template–primer substrate, initiation of reverse transcription involves binding of dsRNA. The current study presents a structure of an RT/dsRNA complex that has the basic components of a reverse transcription initiation complex (RTIC). The unique structural features help understand the significantly slower rate of nucleotide incorporation by an RTIC compared with a catalytically efficient reverse transcription elongation complex.
