Parkinson s disease is a common, disabling brain disorder in which neurodegeneration leads to a variety of motoric and non-motoric symptoms. Loss-of-function mutations in PARK2, the gene encoding theE3 ubiquitin ligase Parkin, are the most frequent cause of recessive Parkinson s disease (PD). Parkin translocates from the cytosol to depolarized mitochondria, ubiquitinates outer mitochondrial membrane proteins and induces selective autophagy of the damaged mitochondria (mitophagy). Parkin may thus be neuroprotective by mediating the removal of dysfunctional mitochondria which would otherwise lead to increased oxidative stress and induction of apoptosis.
