The highly conserved bacterial homospermidine synthase (HSS) is a key enzyme of the polyamine metabolism of many α-, γ-, and δ-proteobacteria and, in particular, of that of pathogenic strains such as Legionella pneumophila and Pseudomonas aeruginosa; HSS might thus serve as a target for new antibiotic drugs. As a model for the bacterial HSS family, various crystal structures of Blastochloris viridis HSS (Bv HSS), including substrate and inhibitor bound structures, have allowed a detailed analysis of an identified deep cavity, including two substrate binding sites at the active site, one of which is highly specific for putrescine.
