The comparison of organon teknika MB/BacT system with Löwenstein-Jenset medium for detection of mycobacteria

Şubat-Ekim 1996 tarihleri arasında Ege Üniversitesi Tıp Fakültesi Mikrobiyoloji ve Klinik Mikrobiyoloji Anabilim Dalı Mikobakteriyoloji Laboratuvar'ında 518 klinik örnek incelemeye alındı. Örnekler, dekontaminasyon uygulandıktan sonra Organon Teknika MB/BactT sistemine ve ayrıca doğrudan Lowenstein-Jensen (LJ) besiyerine inoküle edildi. Direkt mikroskobik incelemeler Kinyoun boyamasıyla yapıldı. Mycobacterium tuberculosis kompleks (MTB) t-anımlamasında biyokimyasal testler kullanıldı. İncelenen 518 örneğin 63'ü (% 12.1) otomatize sistem ile, 36'sı (% 66.9) ise doğrudan LJ besiyerine yapılan kültürler ile pozitif olarak saptandı. MB/BacT sistemi tarafından pozitif olarak değerlendirilen 63 kültürün 55'i (% 87.3) MTB ve diğer sekizi (% 12.6) M spp olarak tanımlanırken; pozitif saptanan 36 LJ kültürünün 24'ü (% 66.6) MTB ve 12'si (% 33.3) de M spp olarak değerlendirildi. Pozitifliğin ortalama saptanma süresi MB/BacT sisteminde MTB için 18.3 gün ve M spp için 20.2 gün olarak saptanırken LJ besiyerlerine yapılan kültürde bu süreler sırası ile 23 ve 28 gün olarak bulundu. Mikobakteri kökenlerinin klinik örneklerden izolasyonunda MB/BacT sistemi LJ besiyeri yöntemine göre daha duyarlı ve hızlı bulundu.Between February and October 1996, 518 clinical specimens were evaluated in the Mycobacteriology Laboratory, Department of Microbiology and Clinical Microbiology, Ege University Hospital. After decontamination, samples were inoculated in Organon Teknika MB/BacT bottles and also directly onto Loewenstein-Jensen (LJ) medium. Direct microscopic examinations were made by Kinyoun stain. For identification of MTB biochemical tests were performed. While 63 of 518 (12.1%) samples were positive by MB/BacT system, 36 (6.9 %) were LJ culture positive. Fifty five of 63 (87.3 %) MB/BacT positive samples were identified as MTB and remaining 8 (12.6 %) were M spp. The median time for MB/Bact system to detect MTB was 18.3 and 20.2 days for M spp. In LJ cultures median detection times were 23 days and 28 days, respectively:- MB/BacT System was found to be a faster and more reliable method than LJ medium for recovery of myçobacteria from clinical specimens
