The Effect of Modified Atmosphere Packaging (MAP) on Spoilage Bacteria inFillets of Live Stored Cod

Forbruket av fersk sjømat øker og forbrukere kjøper torsk oftere enn før, gjerne i porsjonspakninger. Fersk torsk er primært en sesongvare, som er ønsket tilgjengelig året rundt. Forskningsprosjektet CATCH jobber med å gjøre dette mulig, ved hjelp av levendelagring av villfanget torsk. Målet med denne oppgaven var å undersøke effekten av to ulike pakkegasser på vekst av kvalitetsforringende bakterier på levendelagret torsk. Torskefilet ble inokulert med bakteriestammer tidligere isolert fra torsk i forbindelse med CATCH- prosjektet. Torskefiletene ble pakket i modifisert atmosfære (MAP) med 60% CO2 + 40% O2 gass eller 60% CO2 + 40% N2 gass og undersøkt gjennom en lagringsperiode på 13 dager, med prøveuttak ved dag 0, 6, 9 og 13. Det er tre bakterier som er identifiser som ansvarlige for forringelsen av fersk torsk: Photobacterium phosphoreum, Pseudomonas spp. og Shewanella putrefaciens. Det ble ikke funnet S. putrefaciens ved bakterieisoleringen, derfor ble ikke bakterien benyttet i oppgaven. Pseudomonas spp. er en aerob bakterie som er følsom mot CO2. Bakterien danner forbindelser som gir fruktig, råtten og svovelliknende lukt og smak. Photobacterium phosphoreum kan vokse under anaerobe forhold og er tolerant ovenfor høye CO2 konsentrasjoner. Bakterien benytter seg av TMAO som terminal elektronakseptor slik at den svært illeluktende forbindelsen TMA dannes, en forbindelse som bidrar til fordervet fiskelukt. Resultatene fra studiet antydet at pakking med 60% CO2 + 40% O2 gass hadde best inhiberende effekt på de forringende bakteriene, og bevarte ferskheten i torskefiletene lengst. Det så ut til at den høye oksygenkonsentrasjonen bidro til å forsinke bakterieveksten og senket dannelsen av TMA i forhold til pakkegass uten oksygen. Ved sensorisk vurdering av lukt var torskefilet i 60% CO2 + 40% O2 gass fortsatt akseptabel etter 13 dagers lagring, mens torskefilet i 60% CO2 + 40% N2 gass utviklet en uakseptabel lukt etter 9 dagers lagring. The consumption of fresh seafood is increasing. Consumers buy cod more often than before, preferably in portion packs. Fresh cod is primarily available in season, but there is a demand to make it available all year around. The research program CATCH is working on making this possible by capturing wild cod and keeping them alive in net pens. The aim of this study was to survey the effect of two different modified atmospheres on the growth of spoilage bacteria on live stored cod. Cod fillets were inoculated with strains of bacteria previously isolated in the CATCH research program. The cod fillets were then packed in modified atmosphere (MAP) either with 60% CO2 + 40% O2 gas or 60% CO2 + 40% N2 gas. The storage trial lasted for 13 days where sampling was done on day 0, 6, 9 and 13. There are three bacteria identified as specific spoilage bacteria of fresh cod: Photobacterium phosphoreum, Pseudomonas spp., and Shewanella putrefaciens. During the isolation of bacteria, no strains of S. putrefaciens were detected and therefore not used in this study. Pseudomonas spp. is an aerobic bacteria which is sensitive against CO2. The bacteria produce compounds which give a fruity, rotten and sulfur like taste and smell. Photobacterium phosphoreum is able to grow under anaerobic conditions and is tolerant to high CO2 concentrations. The bacteria can use TMAO as a terminal electron acceptor which results in the formation of TMA, a compound that is associated with the smell of spoiled fish. The results from this study indicates that the atmosphere containing 60% CO2 + 40% O2 gas had the best inhibition effect on the spoilage bacteria and the ability to preserve the fish freshness throughout the storage period. It looked like the high oxygen concentration helped in the retardation of the spoilage bacteria and also lowered the formation of TMA. A sensory evaluation of the smell from the cod fillets gave better results for the fillets packed in an atmosphere with oxygen compared to the atmosphere without oxygen. Filets packed in 60% CO2 + 40% O2 gas still had a acceptable smell after 13 days of storage, while filets packed in 60% CO2 + 40% N2 gas had an unacceptable smell at this point of the storage trial.M-MA
