In order to evaluate the effects of space radiation to germ line cells and embryos, we irradiated mouse ES cells and the sell survival and their development was examined. Mouse cultured and frozen ES cells (-80 degree in C) were irradiated by X-ray, Fe and C ion heavy particles. The cells were plated and numbers of colony were counted. The mouse ES cells were most sensitive to Fe ion particles at 0-1Gy of exposure and carbon gave moderate effect to ES cells. Interestingly, radiation at low dose of 0-1 Gy, by Fe and C particles damaged ES cells seriously, while X-ray did not We also examined phosphorylation of histone H2AX after irradiation of X-ray, Fe particles.
