Cambios en la citología endometrial durante el ciclo estral en vacas de tambo

The cytobrush (CB) has proved to be the best technique for obtaining uterine cytology for the diagnosis of subclinical endometritis (SE) in postpartum dairy cows. The diagnosis is based on assessing the percentage of neutrophil cells (%PMNN) in an endometrial cytology smear according to days postpartum. Several authors have described a physiological increase of neutrophils (PMNN) in the endometrium, from late proestrus to metestrus. This increase could eventually alter the diagnosis of ES, resulting in false positive diagnoses. The aim of this study was to evaluate the variation of percentage of PMNN, mononuclear leukocytes cells and the size of endometrial cells in clinically healthy synchronized cows. For the experiment, Holstein dairy cows between 27-56 days postpartum (dpp) at the beginning of the sampling period were used. The cows were synchronized with an Ovsynch protocol and were sampled on days 0, 4, 11 and 18 of the estrous cycle (endometrial cytology for SE diagnosis and blood for progesterone measurement). Data obtained showed that %PMNN did not change throughout the estrous cycle (1.51± 0.50, P> 0.64). These results demonstrate that the %PMNN does not vary along the estrous cycle, and therefore diagnosis of ES by CB technique reflects the inflammatory changes occurred in the endometrium. El diagnóstico se basa en el porcentaje de polimorfo nucleares neutrófilos (%PMNN) encontrados en la citología endometrial según los días postparto en que se encuentra la hembra. Sin embargo, varios autores han descripto un aumento fisiológico de polimorfo nucleares neutrófilos (PMNN) en el endometrio, entre fin del proestro a fines del metaestro, que podría llegar a alterar el diagnóstico de ES resultando en diagnósticos falsos positivos. El objetivo de este estudio fue evaluar a través de citología endometrial, las variaciones de los porcentajes de PMNN, de leucocitos mononucleares y del tamaño de las células endometriales a lo largo del ciclo estral en vacas clínicamente sanas y con celos sincronizados. Se utilizaron vacas Holstein cíclicas (n= 17) de 27-56 días postparto (dpp) al comienzo del muestreo; fueron sincronizadas mediante un protocolo de Ovsynch y se las muestreó a los días 0, 4, 11 y 18 del ciclo (citología endometrial y sangre para medición de las concentraciones séricas de progesterona). Los resultados encontrados mostraron que no hay variaciones significativas por lo que el %PMNN no varía a lo largo del ciclo estral (1,51± 0,50, P> 0,64), y por lo tanto, el diagnóstico de ES por la técnica de CB refleja los cambios inflamatorios que ocurren en el endometrio.
