Establecimiento y multiplicación de suspensiones celulares de Cymbopogon citratus (D.C) Stapf

Cellular suspensions settled down starting from callus of Cymbopogon citratus (D.C) Stapf cultivated in semisolid medium, according to the methodology described for Freire (1998), for the cultivation of the cane of sugar and later on modified by Licea and Gómez (2000) for the cultivated callus of Cane Santa, with the objective of analyzing the effect of the cellular density on the cellular growth, being studied the behavior of the fresh mass, dry mass and the pH in three inocule densities (20, 40 and 60 gMF.l-1). The development of roots was evaluated in the cellular aggregated and it was also analyzed directly the influence of the explants on the callus formation cultivated directly in liquid medium, starting from cultivated plants in vitro. The biggest increment of fresh mass was obtained when 20 gMF.l-1 was used, the values of mass dry off they behaved in a similar way, being obtained the biggest rate of growth in this same treatment. The pH in the three densities of studied inocule, diminished during the first eight days and stayed stable starting from this moment. The alone presence of roots was appreciated only in the cellular aggregated cultivated without coconut water. The formation of callus directly in liquid medium took place in the region near to the meristematic area.Key words: coconut water, biomass production, lemon grass, root formationSe establecieron suspensiones celulares a partir de callos cultivados en medio de cultivo semisólido de Cymbopogon citratus (D.C) Stapf, según la metodología descrita por Freire (1998), para el cultivo de la caña de azúcar y posteriormente modificada por Licea y Gómez (2000) para el cultivo de callos de Caña Santa. Con el objetivo de analizar el efecto de la densidad celular sobre el crecimiento celular, se estudió el comportamiento de la masa fresca, masa seca y el pH en tres densidades de inóculo (20, 40 y 60 gMF.l-1). Se evaluó el desarrollo de raíces en los agregados celulares y se analizó además la influencia del explante sobre la formación de callos directamente en medio de cultivo líquido, a partir de plantas cultivadas in vitro. El mayor incremento de masa fresca se obtuvo cuando se empleó 20 gMF.l-1. Los valores de masa seca se comportaron de manera similar, obteniéndose la mayor tasa de crecimiento en este mismo tratamiento. El pH en las tres densidades de inóculo estudiadas, disminuyó durante los primeros ocho días y se mantuvo estable a partir de este momento. La presencia de raíces solo se apreció en los agregados celulares cultivados en medio de cultivo sin agua de coco. La formación de callos directamente en medio de cultivo líquido solo tuvo lugar en el cilindro de la vaina de las hojas más próximo a la zona meristemática.Palabras clave: agua de coco, Caña Santa, formación de raíces, producción de biomas
