The objective of this project was to transform the diatom P. tricornutum by electroporation in order to optimize the method. A test protocol was composed in order to incorporate zeocin resistant pPha-T1-GFP plasmid, linearized by ScaI-HF digestion. Transformation of P. tricornutum with electroporation is not entirely new nor is it common either due to poor yield of transformants. Electroporation is a method that offers simple solutions, especially if it’s combined with a successful optimizing protocol.
