PENETAPAN KADAR TOKOFEROL MINYAK BUAH MERAH (Pandanus conoideus Lam.) SECARA IN VITRO DAN IN VIVO PADA TIKUS YANG DIBERI BEBAN AKTIVITAS FISIK MAKSIMAL

ABSTRACTExogenous antioxidant is required to prevent the occurrence of oxidative stress that can make cell damage.Tocopherol content of red fruit oil is thought to have biological activity as antioxidants. Tocopherol effectivelyprevent the lipid peroxidation and free radical formation. The objective of the study was to evaluate quantitaveconcentration and the effect of buah merah oil to tocopherol level in blood during maximum physical activitytreatment. Tocopherol concentration of buah merah oil in vitro was measured using Abe and Katsui methode byfluorometer. Antioxidant activity in vivo test was done using 24 male Wistar rats in pre-post test control groupdesign. The rats were grouped into 4 group. The control group was given destilated water and three treatmentgroups were given buah merah oil in the dose of 0,15 ml/kg bw; 0,3 ml/kg bw; 0,6 ml/kg bw for 10 days. Beforebuah merah oil was given, level of tocopherol was measured. Ten days later, the four group were given maximumphysical activity mean of swimming until the sign of fatigue occurred (nearly drowned) and the blood was takenfor blood tocopherol examination. Tocopherol concentration of buah merah oil was 447μg/ml. In vivo test, dosage0,15 ml/kg bw could increase tocopherol 15,46%. Dosage 0,3 ml/kg bw could increase tocopherol 22,19% anddosage 0,6 ml/kg bw could increase tocopherol 50,60%. In vitro, tocopherol concentration of buah merah oil was447μg/ml. In vivo experiment, all dosage of buah merah oil increased the blood tocopherol level.ABSTRAKPemanfaatan senyawa antioksidan eksogen secara efektif sangat diperlukan untuk mencegah terjadinya stresoksidatif yang berakibat pada kerusakan sel. Kandungan tokoferol dalam minyak buah merah tinggi dandiduga memiliki aktivitas biologi sebagai antioksidan. Tokoferol efektif mencegah terjadinya peroksidasi lipiddan pembentukan radikal bebas. Penelitian ini bertujuan untuk menguji kandungan tokoferol serta mengkajipengaruh minyak buah merah terhadap kadar tokoferol dalam darah pada pemberian aktifitas fisik maksimal.Kadar tokoferol dalam minyak buah merah in vitro diukur dengan metode Abe dan Katsui secara fluorometri.Uji aktivitas antioksidan secara in vivo dilakukan dengan menggunakan 24 ekor tikus putih galur Wistarjantan dengan pre-post test control group design. Tikus dibagi dalam 4 kelompok. Pada kelompok 1 sebagaikelompok kontrol diberi akuades dan tiga kelompok pelakuan yang masing-masing diberi minyak buah merahdosis 0,15 ml/kg bb; 0,3 ml/kg bb; 0,6 ml/kg bb selama 10 hari. Sebelum pemberian minyak buah merah, dilakukan pengukuran kadar tokoferol pada semua kelompok. Pada hari ke-10, keempat kelompok diberikan beban aktivitas fisik maksimal yaitu dibuat berenang sampai kelelahan berupa hampir tenggelam dan segeradilakukan pengambilan darah untuk mengukur kadar tokoferol dalam darah. Pada uji in vitro, kandungantokoferol pada minyak buah merah adalah 447 μg/ml. Pada uji in vivo dengan dosis 0,15 ml/kg bb, kadartokoferol meningkat 15,46%. Pada dosis 0,3 m/kg bb, kadar tokoferol meningkat 22,19% dan pada dosis 0,6ml/kg bb, kadar tokoferol meningkat 50,60%. Kandungan tokoferol pada minyak buah merah adalah 447 μg/ml.
