Efek Ekstrak etanol Buah Flagellaria indica L. Terhadap Kualitas Sperma dan Penampilan Histologi Tubulus Seminiferus mencit Swiss Webster albino

ABSTRACT Kahayan Hilir people is being used Flagelaria indica L (Flagellariaceae) fruit as traditional fertility control. In-terestingly it fruit used as antifertility agent in male and female. Although the plant is a well known antifertility plant, till today poorly knows of male antifertility activity of it fruit. The present study was to evaluate the effect of antifertility in ethanol extract of Flagelaria indica L. fruit has been studied in Albino mice. The study was divided into four groups of six animals for each. The first group (I) received sterile aquabidest serve as control. The second and third groups (II, III and IV) of animals were gavage the ethanol extract of Flagelaria indica L. fruit on 0,021 mg/kg body wt., 0,042 mg/kg body wt., and 0,084 mg/kg body wt respectively for a period of three weeks. Mice kill at 21th days the animals were sacrificed by dislocation. Sperm collected from vas deferens, motility, number of normal and abnormal sperm were count. Testis was isolated, washed in buffered saline and fixed in Bouin’s solu-tion. Testis sections (5-6 μm) were routinely processed by standard histological techniques, stained with hematox-ylin and eosin (HE), and examined by light microscope (Nikon Eclipse E400) to assess structure changes among control and experimental animals. The result shows testes of mice group treated with the extract dose 0.021 mg/ kg bw cause seminiferous tubules wall becomes thinner, the lumen becomes wider, and number of sperm in the lumen decline. At doses of 0.042 mg/kg bw and 0.084 mg/kg semiferus tubule wall disorganize, but interstitial cell dtructure normal. Conclusion: ethanol extract of Flagelaria indica L. fruit causes a decrease in sperm quality and disorganization of the seminiferous tubule, thereby disrupting spermatogenesis.AbstrakPenduduk Kahayan Hilir dan sebagian penduduk propinsi Kalimantan Tengah menggunakan buah Flagelaria indica L. (Flagellariaceae) untuk mengendalikan fertilitas. Keunikannya, buah tumbuhan tersebut tidak hanya digunakan untuk oleh wanita maupun pria. Walaupun buah tumbuhan ini telah diketahui khasiatnya sebagai agensia antifertilitas, namun mekanisme kerjanya sebagai aghen antifertilitas pria belum diketahui. Pada pe-nelitian ini digunakan empat kelompok yang masing-masing terdiri enam mencit. Kelompok I sebagai control digavage akuabides steril, sedangkan kelompok II, III dan IV masing-masing digavage dengan ekstrak etanol buah F. indica L. dengan dosis 0,021 mg/kb bb, 0,042 mg/kg bb dan 0,084 mg/kg bb. Penelitian dilakukan selama 3 minggu. Pada hari ke 21 mencit dibunuh dengan cara dislokasi leher, kemudian sperma dari vas deferen diambil dan diamati motilitas, jumlah sperma nomal dan jumlah sperma abnormal. Testis diisolasi, kemudian dicuci dengan garam fisiologis dan difiksasi dalam larutan bouin. Testis disayat pada ketebalan 5-6 µm dan diwarna menggunakan hematoksilin Eosin (HE), dan diamati menggunakan mikroskup Nikon eclipse E400. Hasil penelitian menunjukkan, ekstrak buah uwei namei menyebabkan penurunan motilitas sperma, penurunan jumlah sperma dan peningkatan prosentase sperma abnormal secara sangat signifikan. Hasil pengamatan sayatan testis mencit kelompok perlakuan dengan dosis ekstrak uwei namei 0,021 mg/kg bb menunjukkan, bahwa ekstrak buah uwei namei menyebabkan dinding tubulus seminiferus menjadi lebih tipis, lumen menjadi lebih lebar, jumlah sperma dalam lumen mulai menurun. Pada dosis yang 0,042 mg/kg bb dan 0,084 mg/kg terjadi kerusakan dinding tubulus semiferus, namun tidak merusak struktur sel interstial. Berdasarkan hasil tersebut disimpulkan ekstrak buah uwei namei menyebabkan penurunan kualitas sperma dan disorganisasi dinding tubulus seminiferus, sehingga mengganggu spermatogenesis
