ABSTRACTDental plaque is a soft deposit forming biofilms attached to the tooth surface and containing Streptococcus mutans.This can lead to a variety of dental and oral diseases especially dental caries. Therefore, the formation ofdental plaque should be prevented, one with rinse using steeping green betel leaf (Piper betle L.). The purposeof this study was to assess the effect of rinsing steeping green betel leaf against dental plaque formation and developmentof the Colony Forming Units (CFU) of Streptococcus mutans in saliva.The design of this research is theexperimental post-test control group. The research subject in this study were 20 people who divided into 2 treatmentgroups (n = 10). Group I gargle with steeping green betel leaf and group II rinsing with sterile distilled water.The measured data is dental plaque index and the number of CFU/ml of Streptococcus mutans in saliva. Datawas analysed by unpaired t test, α = 0,05.The results of the study showed that mean plaque index in group I afterrinsing with steeping green betel leaf was 31,94 %, highly significantly different (p < 0,01) compared to group IIafter rinsing with sterile distilled water for 73,60 %. Colony Forming Units/ml in saliva after rinsing with steepinggreen betel leaves was 6.28, andit is less than after rinsing sterile distilled water (7,94), with a highly significantdifference (p < 0,01).The conclusions of this study is rinsing with steeping green betel leaf may reduce dentalplaque index and lower the number of Colony Forming Units bacterium Streptococcus mutans in the oral cavity. Hal ini dapat menyebabkan terjadinya berbagai penyakit gigi dan mulut, terutamakaries gigi. Oleh karena itu pembentukan plak gigi harus dicegah, salah satunya dengan berkumur menggunakanseduhan daun sirih hijau (Piper betle L.). Tujuan dari penelitian ini adalah untuk menilai efek berkumurseduhan daun sirih hijau (Piper betle L.) terhadap pembentukan plak gigi dan perkembangan Colony FormingUnit (CFU) Streptococcus mutans pada saliva.Desain penelitian ini adalah eksperimental sungguhan denganpost test only control group. Naracoba pada penelitian ini berjumlah 20 orang yang dibagi menjadi 2 kelompokperlakuan (n=10). Kelompok I berkumur dengan seduhan daun sirih hijau (Piper betle L.) dan kelompok IIberkumur dengan aquadest steril. Data yang diukur adalah indeks plak gigi dan jumlah CFU/ml Streptococcusmutans pada saliva. Analisis data dengan uji T tidak berpasangan, α=0,05. Hasil penelitian rerata indeks plakgigi pada kelompok I setelah berkumur dengan seduhan daun sirih hijau (Piper betle L.) (31,94 %), berbeda sangat bermakna (p<0,01) dibandingkan dengan kelompok II setelah berkumur dengan aquadest steril sebesar(73,60 %). Colony Forming Unit/ml pada saliva setelah berkumur dengan seduhan air sirih hijau (Piper betleL.) (6,28), lebih sedikit dibandingkan dengan setelah berkumur aquadest steril (7,94), dengan perbedaan yangsangat bermakna (p<0,01). Berkumur dengan seduhan daun sirih hijau (Piper betle L.) dapat menurunkanindeks plak gigi dan menurunkan jumlah Colony Forming Unit bakteri Streptococcus mutans di rongga mulut
