Librería sustractiva obtenida de un mutante de la variedad B 4362 con resistencia a la roya de la caña de azúcar

The hypersensitive response is one of the most powerful mechanisms for which the plants resist pathogen attack. Mutations carried out previously on the variety B4362, of sugarcane, originated five mutants that express this mechanism towards the attack of rust (Puccinia melanocephala Syd.). By means of a subtractive hybridization among the cDNA obtained starting from the resistant clone inoculated with rust and a pool of cDNA of the susceptible variety (B4362) inoculated and of the resistant clone not inoculated, it was possible to reduce the number of genes expressed during the infection with the fungus. A subtractive library was carried out where we hope that most of the genes are involved in the hypersensitive response that present these mutants towards the infection of the pathogen.Key words: Subtractive hybridization, hypersensitive response, Puccinia melanocephala Syd.La respuesta hipersensible es uno de los mecanismos más poderosos por el cual las plantas resisten el ataque de patógenos. Mutaciones realizadas, previamente, sobre la variedad B4362, de caña de azúcar, originaron cinco mutantes que expresan este mecanismo frente al ataque de la roya (Puccinia melanocephala Syd.). Mediante una hibridación sustractiva entre el cADN obtenido a partir del clon resistente inoculado con roya y un pool de cADN de la variedad susceptible (B4362) inoculado y del clon resistente no inoculado, se logró reducir el número de genes expresados durante la infección con el hongo. Se realizó una librería sustractiva donde se espera se encuentren la mayoría de los genes involucrados en la respuesta de hipersensibilidad que presentan estos mutantes frente a la infección del patógeno.Palabras clave: hibridación sustractiva, Puccinia melanocephala Syd, respuesta hipersensibl
